Coxiella burnetii is een unieke intracellulaire ziekteverwekker die de zoönotische menselijke besmetting Q-koorts veroorzaakt. De BLAM substraat bestaat uit twee fluoroforen (coumarine en fluoresceïne), die een FRET-paar vormen. Deze translocatie assay is bewezen als een benadering van effector eiwitten te identificeren van een reeks verschillende intracellulaire en extracellulaire bacteriën, waaronder C. burnetii, L. pneumophila, L. longbeachae, Chlamydia trachomatis, enteropathogene E. coli, Salmonella en Brucella 17,30-35. Om de rol van specifieke host factoren op Coxiella effector translocatie bepalen we gebruik van een bekende methode voor het gene silencing bekend als RNA-interferentie, in het bijzonder kleine interfererende RNA (siRNA). Oorspronkelijk geïdentificeerd in Caenorhabditis elegans, RNA-interferentie is een geconserveerd endogeen cellulair proces dat wordt gebruikt voor het aangeboren defense tegen virussen en genregulatie 36,37. Na binding van sequentie-specifieke siRNA, afbraak van mRNA vindt plaats via RISC (RNA-geïnduceerd silencing complex) waardoor specifiek gen silencing of 38 knockdown. In deze studie werd gebruikt siRNA targeten twee gastheereiwitten, Rab5A en Rab7A, die belangrijke regulatoren van de endocytische route zijn.
