Klassiska monocyter uttrycker en hög nivå av CD14 men uttrycker inte CD16 (CD14 ++ CD16 -), mellan monocyter uttrycker en hög nivå av CD14 och en mellannivå av CD16 (CD14 ++ CD16 +), och icke-klassiska monocyter uttrycker en låg nivå av CD14 och en hög nivå av CD16 (CD14 + CD16 ++). Monocyter som uttrycker CD16 betecknas CD16 + monocyter, som jämfört med CD16 - monocyter har hög expression av inflammatoriska cytokiner och förmågan att mer effektivt presentera antigener 13,14. Ungefär 10% av monocyter uttrycker CD16 under homeostas med högre procenttal som observerats under inflammation 15. Monocyt subpopulationer är förknippade med vissa sjukdomstillstånd och kan vara användbara biologiska markörer för sjukdom och sjukdomsutveckling 16. Vårt mål var att identifiera en metod som kan mäta glukostransportör uttryck och glukosupptag av humana monocyter och monocyt subpopulationer under förhållanden så nära physiological förhållanden som möjligt. Tidigare studier mätt monocyt glukostransportör uttryck och glukosupptag 17,18, men dessa metoder undersökte isolerade monocyter som kan ha förändrat proteinuttryck jämfört med fysiologiska betingelser 19, och ingen tidigare studie har undersökt humana monocyt subpopulationer. Monocyter är medfödda immunceller som kan aktiveras av patogener och inflammation i samband med vissa kroniska inflammatoriska sjukdomar. Aktivering av monocyter inducerar effektorfunktioner och en åtföljande övergång från oxidativ till glykolytiska metabolismen som är förenad med ökad glukostransportör uttryck. Denna ökade glykolytiska metabolismen observeras även för utbildad immunitet monocyter, en form av medfödd immunologiskt minne. Även in vitro protokoll undersöker glukostransportör uttryck och glukosupptag av monocyter har beskrivits, har ingen granskats av flera parametrisk flödescytometri i helblod. Vi beskriver en multi-parametrisk flödescytometrisk protokoll för mätning av fluorescerande glukos analog 2-NBDG upptag i helblod med totala monocyter och den klassiska (CD14 ++ CD16 -), mellan (CD14 ++ CD16 +) och icke-klassiska ( CD14 + CD16 ++) monocytsubpopulationer. Denna metod kan användas för att undersöka glukostransportör uttryck och glukosupptag för total monocyter och monocyt subpopulationer under homeostas och inflammatorisk sjukdom, och kan lätt modifieras för att undersöka glukosupptag för andra leukocyter och leukocyter subpopulationer inom blod.

Ersättning måste utföras för enskilda fluorokromer för att förhindra fluorescens spridningseffekter. Monocyter först berikas av grindning baserat på framåt och sidospridning.
