microfluidic buffer exchange for interference-free micro/nanoparticle cell engineering

cellules d&#39;ingénierie avec micro ingrédient actif chargé / nanoparticules (IP) devient une méthode de plus en plus populaire pour améliorer les propriétés thérapeutiques indigènes, activer bio imagerie et contrôler le phénotype cellulaire. Une question cruciale encore insuffisamment abordée est le nombre important de particules qui restent non liés après marquage des cellules qui ne peuvent pas être facilement éliminé par centrifugation classique. Cela conduit à une augmentation de la bio bruit imagerie de fond et peut donner des effets transformateurs sur des cellules non-cibles voisines. Dans ce protocole, nous présentons un tampon stratégie d&#39;échange inertiel à base microfluidique appelé comme Dean flux Fractionnement (DFF) pour séparer efficacement les cellules marquées de NPs libres d&#39;une manière à haut débit. Le microdispositif spirale développé facilite la collecte continue (&gt; de récupération des cellules de 90%) des cellules purifiées (THP-1 et CSM) en suspension dans une nouvelle solution tampon, tout en atteignant&gt; 95% épuisement de colorant fluorescent non lié ou NPs colorant chargé (silique ou PLGA). Cette seule étape, la stratégie de purification de cellules basée sur la taille permet de haute cellule de traitement débit (10 6 cellules / min) et est très utile pour un grand volume de purification de cellules de micro cellules / nanoparticulaires conçue pour parvenir à une application clinique sans interférence.

Après marquage des cellules avec bio imagerie IP de l&#39; agent chargé d&#39;une nuit, les cellules marquées (contenant des particules libres) sont récoltées et purifiées par FFD spirale microdispositif pour éliminer les IP libres dans un processus en une seule étape (figure 1A).
