células de ingeniería con cargadas con sustancias activas, micro / nanopartículas (NP) se está convirtiendo en un método cada vez más popular para mejorar las propiedades terapéuticas nativas, permitirá bio imágenes y controlar el fenotipo celular. Una cuestión fundamental todavía insuficientemente reconocidos es el importante número de partículas que permanecen sin unir después del marcaje de células que no se pueden eliminar fácilmente mediante centrifugación convencional. Esto conduce a un aumento en el ruido de fondo de imagen bio y puede impartir efectos transformadores en las células no objetivo vecinos. En este protocolo, se presenta una estrategia basada en la microfluídica inercial tampón de intercambio denominado como Decano de flujo de fraccionamiento (DFF) para separar eficientemente las células marcadas de PN libres en una forma de alto rendimiento. El microdispositivo espiral desarrollado facilita la recogida continua (&gt; recuperación de células 90%) de células purificadas (THP-1 y MSC) en suspensión en una nueva solución tampón, mientras que el logro% agotamiento de colorante fluorescente no unido o NPs de tinte cargado (sil&gt; 95ica o PLGA). Esto solo paso, la estrategia de purificación de células basadas en el tamaño hace posible un rendimiento de procesamiento de alta celular (10 6 células / min) y es muy útil para la purificación de células a gran volumen de células micro / nanopartículas de ingeniería para lograr la aplicación clínica libre de interferencias.
