I detta arbete, beskriver vi en okomplicerad, snabb film dispersion metod för framställning av miceller. Kemoterapi används ofta för att behandla olika former av cancer. De flesta, om inte alla, cytostatika har toxiska biverkningar som kan variera från hanterbara mindre förhållanden, såsom illamående och diarré, mer livshotande tillstånd. Eftersom de flesta läkemedel mot cancer är giftiga, icke-selektiva exponeringen av dessa läkemedel till normal vävnad orsakar oundvikligen toxicitet. Det finns därför ett stort behov av ett terapeutiskt angreppssätt som selektivt kan leverera läkemedel in i cancerceller. En annan utmaning med administrering av läkemedel mot cancer är deras dåliga vattenlöslighet. Vanligtvis är solubiliseringsmedel som behövs för att formulera dessa svårlösliga läkemedel. Dock kan de flesta solubiliseringsmedel, såsom dimetylsulfoxid (DMSO), Cremophor EL, och polysorbat 80 (Tween 80) orsaka lever och njurtoxicitet, hemolys, akuta överkänslighetsreaktioner och perifera neuropatier. 1 Därför är säkra och biokompatibla formuleringar som behövs för den kliniska användningen av dåligly lösliga cytostatika. Nanocarriers är lovande läkemedelsleveranssystem för att ta itu med dessa utmaningar. Doxil, Abraxane, och Genexol) har godkänts av tillsynsmyndigheter för behandling av cancerpatienter. 10 Polymera miceller är lovande nanoskala drug delivery bärare, som har framgångsrikt använts för leverans av läkemedel mot cancer. 4-7,11,12 Typiska polymera miceller framställs från amfifila polymerer genom en självorganiserande processen. Kärna-skal-strukturerade polymera miceller innefattar ett hydrofilt skal och en hydrofob kärna. Den hydrofila skalet kan steriskt stabilisera miceller och förlänga deras omsättning i blodet. Den hydrofoba kärnan effektivt kan kapsla hydrofoba dmattor. På grund av den lilla storleken av miceller (typiskt mindre än 200 nm) och långcirkulations egenskaper, polymera miceller tros uppnå tumör inriktning genom ökad permeabilitet och retention (EPR) effekter (passiv tumörmåls). Nyligen har mycket arbete gjorts för att förbättra laddningen av läkemedel i leveransbärare. Den lipofila prodrug strategi är en strategi som har undersökts för att förbättra läkemedelsbelastning i lipid nanopartiklar och emulsioner. 13,14 den jämfördaugation av lipider med läkemedel kan avsevärt förbättra sin lipofilicitet och förbättra lastning och retention i de lipofila komponenterna i nanocarriers. Här beskriver vi ett protokoll för framställning av lipofila doxorubicin pro-läkemedelsladdade miceller. För det första är syntesförfarande för doxorubicin lipofila pro-läkemedel beskrivits. preparation and characterization of lipophilic doxorubicin pro-drug micelles

Micelles have been successfully used for the delivery of anticancer drugs. Amphiphilic polymers form core-shell structured micelles in an aqueous environment through self-assembly. The hydrophobic core of micelles functions as a drug reservoir and encapsulates hydrophobic drugs. The hydrophilic shell prevents the aggregation of micelles and also prolongs their systemic circulation in vivo. In this protocol, we describe a method to synthesize a doxorubicin lipophilic pro-drug, doxorubicin-palmitic acid (DOX-PA), which will enhance drug loading into micelles. A pH-sensitive hydrazone linker was used to conjugate doxorubicin with the lipid, which facilitates the release of free doxorubicin inside cancer cells. Synthesized DOX-PA was purified with a silica gel column using dichloromethane/methanol as the eluent. Purified DOX-PA was analyzed with thin layer chromatography (TLC) and 1H-Nuclear Magnetic Resonance Spectroscopy (1H-NMR). A film dispersion method was used to prepare DOX-PA loaded DSPE-PEG micelles. In addition, several methods for characterizing micelle formulations are described, including determination of DOX-PA concentration and encapsulation efficiency, measurement of particle size and distribution, and assessment of in vitro anticancer activities. This protocol provides useful information regarding the preparation and characterization of drug-loaded micelles and thus will facilitate the research and development of novel micelle-based cancer nanomedicines.
