Dessa mänskliga microbiomes spelar en nyckelroll i metaboliserande näringsämnen och xenobiotika, reglering av immunsvar, och upprätthålla tarm homeostas 2. Inte överraskande, med tanke på dessa olika funktioner, kommen tarmen microbiome modulerar omfattande hälsa och sjukdom 3. Således, förstå betydelsen av tarm microbiome och värd-mikrob-interaktioner är av stor betydelse för att främja gastrointestinal (GI) hälsa och utforska nya läkemedel för tarmsjukdomar 4. Emellertid befintliga in vitro tarmmodeller (t.ex., statiska kulturer) begränsar värd-microbiome sam-kultur till en kort tidsperiod (&lt;1 dag) eftersom mikrobiella celler växa över och äventyra tarmbarriärenfunktion 5. Surrogatdjurmodeller (t.ex. bakteriefri sex eller genetiskt modifierade möss 7) är inte heller vanligt förekommande för att studera värd gut microbiome överhörning eftersom koloniseringen och stabilt upprätthållande av människans tarm microbiome är svåra. För att övervinna dessa utmaningar, nyligen utvecklade vi en biomimetisk människa &quot;Gut-on-a-chip&quot; microphysiological systemet (Figur 1A, vänster) för att efterlikna de värd gut microbiome samspel som sker i den levande mänskliga tarmen 5,8. Här beskriver vi ett protokoll för att utföra långsiktiga samodling av flera arter människans tarm microbiome med microengineered tarmludd i människans tarm-on-a-chip microphysiological enhet. Vi sammanfatta tarm lumen-kapillära vävnadsgränssnittet i en mikroflödessystem enhet, där fysiologiska mekaniska deformationer och fluidfriktionsflöde ständigt tillämpas för att efterlikna peristaltiken. I lumen mikrokanal, humana intestinala epiteliala Caco-2-celler odlas för att bilda en &#39;bakteriefri&#39; villus epitel och regenerera tunntarms villi. Pre-odlade mikrobiella cellerna inokuleras in i lumen sidan för att etablera en värd mikrob ekosystemet. Efter mikrobiella cellerna vidhäftar till den apikala ytan av villi, är vätskeflöde och mekaniska deformationer återupptogs för att producera en steady-state mikromiljö i vilken färskt odlingsmedium konstant matas och obundna bakterier (såväl som bakteriella avfall) kontinuerligt avlägsnas. Efter förlängd samodling from dagar till veckor, har flera mikrokolonier befunnits vara slumpmässigt placerad mellan villi, och både mikrobiella och epitelceller förbli livskraftig och funktionell för åtminstone en vecka i kultur. Vår samodling protokollet kan anpassas för att tillhandahålla en mångsidig plattform för andra värd-microbiome ekosystem som kan hittas i olika mänskliga organ, som kan underlätta in vitro-studie av den roll som humant microbiome i orchestrating hälsa och sjukdom.

Att efterlikna den mänskliga tarmen värd microbiome ekosystem in vitro, är det nödvändigt att utveckla ett experimentellt protokoll för att rekonstruera långsiktigt stabil samodling av tarmbakterier och mänskliga intestinala epitelceller under fysiologiska förhållanden såsom peristaltiken-liknande mekanik och vätskeflöde.
