Il nostro obiettivo per questo studio è quello di consentire la generazione rapida e efficiente del knockout genici mirati in Y. lipolytica ceppo Po1g. A differenza di Saccharomyces cerevisiae, Yarrowia lipolytica, un lievito non convenzionale, può crescere in forma di lievito o micelio in risposta ai cambiamenti delle condizioni ambientali 1,2. Così, questo lievito dimorphic può essere usato come un buon modello per lo studio della differenziazione fungine, morfogenesi e tassonomia 3,4,5. E &#39;generalmente considerato come una specie di lievito sicuri (GRAS), che è ampiamente utilizzato per produrre una varietà di additivi alimentari, come acidi organici, polialcoli, composti aromatici, emulsionanti e tensioattivi 6,7,8,9. È un aerobio obbligato e un lievito oleaginosa noto in grado di accumulare naturalmente lipidi in quantità elevate, cioè, fino al 70% del peso secco cellulare 10. Yarrowia lipolytica è una specie di lieviti non patogeni, dimorfe e rigorosamente aerobici. Grazie alle sue caratteristiche fisiologiche distintive e caratteristiche metaboliche, questo lievito non convenzionale non è solo un buon modello per lo studio della natura fondamentale di differenziazione fungina, ma è anche una piattaforma microbica promettente per la produzione biochimica e varie applicazioni biotecnologiche, che richiedono ampie manipolazioni genetiche. Tuttavia, le manipolazioni genetiche di Y. lipolytica stato limitato a causa della mancanza di un sistema di trasformazione genetica efficiente e stabile, così come molto elevati tassi di ricombinazione non omologa che può essere attribuito principalmente al gene KU70. Qui riportiamo un protocollo semplice e rapido per la trasformazione genetica efficiente e per delezione di un gene in Y. lipolytica Po1g. Innanzitutto, un protocollo per la trasformazione efficiente di DNA esogeno in Y. lipolytica Po1g è stato stabilito. secoND, per ottenere il doppio attraversamento tasso di ricombinazione omologa migliorato per l&#39;ulteriore eliminazione di geni bersaglio, il gene KU70 stato eliminato trasformando una cassetta perturbazione che trasporta 1 braccia kb di omologia. In terzo luogo, per dimostrare il gene maggiore efficienza l&#39;eliminazione dopo la cancellazione del gene KU70, abbiamo eliminato singolarmente 11 geni bersaglio che codificano alcol deidrogenasi e alcool ossidasi con le stesse modalità del ceppo piattaforma di eliminazione diretta KU70. È stato osservato che il tasso di preciso ricombinazione omologa notevolmente aumentato da meno del 0,5% per l&#39;eliminazione del gene KU70 in Po1g al 33% -71% per il singolo gene cancellazione dei 11 geni bersaglio in Po1g KU70 Δ. Un plasmide replicativo portando il marcatore di resistenza igromicina B e il sistema Cre / loxP è stato costruito, e il gene marcatore di selezione dei ceppi di lievito knockout finalmente è stato rimosso con espressione di Cre ricombinasi per facilitare vari cicli di targeted manipolazioni genetiche.
