Veel grote cellulaire processen worden uitgevoerd door grote eiwit en eiwit-RNA-complexen 1 uitgevoerd. Het isoleren van een complex van een natieve bron heeft vele voordelen, waaronder behoud relevante post-transcriptionele en / of translationele modificaties van subeenheden en verzekeren goede complex samenstel. Echter grote cellulaire complexen zijn vaak aanwezig in een cel op een klein aantal kopieën en de zuivering zeer efficiënt moeten zijn en optreden onder fysiologische omstandigheden omgeving te waarborgen complex behouden blijft. Zuiveren van een complex van een eukaryote bron is bijzonder uitdagend en financieel kan worden onbetaalbaar. Een strategie die issuccesvol in het zuiveren van inheemse complexen van eukaryote cellen voor hun eerste karakterisering is de Tandem Affinity Purification (TAP) methode 2,3. TAP methode werd aanvankelijk ontwikkeld met een natief eiwit van gist (S. cerevisiae) zuiveren complex samenwerkende factor (en) 2. TAP methode gebruikt twee labels, elke tag gefuseerd in tandem om hetzelfde eiwit coderende gensequentie. De labels werden geselecteerd om een ​​behoefte voor kleine en selectieve binding aan een affiniteitshars met actieve handhaven dichtbij fysiologische oplossing in evenwicht te brengen. Het genomisch opgenomen TAP tag aan het uiteinde (C-eindstandige) van een eiwit coderend gen en bestond uit een sequentie die codeert voor een calmoduline-bindend peptide (CBP), gevolgd door Proteïne A - toevoeging van iets meer dan 20 kDa gelabelde eiwit. CBP is kort, 26 aminozuren, en door de ~ 17 kDa eiwit calmoduline (CaM) in aanwezigheid van calcium met een Ko in de orde van 10 -9 M 4. Dit is een noodzakelijke stap de interactie van IgG en proteïne A is zeer sterk en kan alleen voldoende worden verstoord onder denaturerende oplossingsomstandigheden. Bij gebrek aan een Protein Een tag wordt het eiwit gebonden CaM hars in aanwezigheid van calcium en geëlueerd uit de hars met toevoeging van het metaalion chelerende EGTA (ethyleenglycol-azijnzuur) (figuur 1). Kort na de invoering van de TAP methode werd in een grootschalig onderzoek gebruikt voor het genereren van een &quot;kaart&quot; van complexe interacties in S. 8 cerevisiae. Belangrijker, als gevolg van deze inspanningen een volledige gist-TAP Tagged open leeskader (ORF) bibliotheek en individuele TAP hebben ORFs 9 kunnen van een commerciële bron. De resolutie verkrijgen van een complex met EM blijft echter afhankelijk van de kwaliteit van het complex onder studie. Deze studie heeft de TAP tag aanpak te zuiveren van S. benut cerevisiae U1 snRNP, een 18-subeenheid (~ 0,8 MDa) laag kopieaantal ribonucleoproteïnecomplex die deel uitmaakt van de spliceosome 15,16. Een aantal maatregelen zijn genomen om dit complex zodanig dat het homogeen en voldoende concentratie zuiveren.
