PMMA-baserte chips er fabrikkert ved hjelp av laser ablasjon og skriving som er billigere og enklere metoder i forhold til PDMS-baserte brikker som krever mer komplisert myk litografi. Etter utforme en microfluidic chip, kan fabrikasjon og montasje gjøres i løpet av bare 5 min. Det er noen viktige skritt at oppmerksomheten bør rettes mot i å utføre eksperimentet. In vivo-celler er omgitt av en rekke av biomolekyler, inkludert ekstracellulære matriks (ECM), karbohydrater, lipider og andre celler. De functionalize ved å svare på mikro-miljø stimuli som interaksjoner med ECM og svar på kjemiske gradienter av ulike vekstfaktorer. Tradisjonelt er in vitro cellestudier utført i cellekulturskåler hvor forbruket av celler og reagenser er stor og cellene vokse i en statisk (ikke-sirkulerende) miljø. Nylig har mikro-fabrikkert enheter integrert med fluidkomponenter gitt en alternativ plattform for cellestudier i en mer kontrollerbar måte. Slike anordninger er i stand til å opprette en nøyaktig mikro-miljø av kjemiske og fysikalske stimuli, mens forbruket av celler og reagenser minimaliseres. Disse microfluidic brikker kan være laget av glass-substrater, silisiumskiver, polydimetylsiloksan (PDMS) polymerer, polymetylmetakrylat (PMMA) substrater, eller polyethylenetereftalat (PET) substrater 1-3. PDMS-baserte enheter er gjennomsiktige, biokompatible, og permeabel for gasser, noe som gjør dem egnet for langtids cellekulturer og studier. PMMA og PET underlag er billig og lett å bli behandlet ved hjelp av laser ablasjon og skriving. Et annet eksempel er å undersøke cellenes retnings migrasjon i henhold elektriske felter (EFS), et fenomen som heter electrotaxis. Electrotactic atferd av celler ble rapportert å være relatert til nerve regenerering 8, embryoutvikling 9,og sårtilheling 10,11. Og mange studier har blitt utført for å undersøke electrotaxis av ulike celletyper, inkludert kreftceller 12,13, lymfocytter 14,15, leukemiceller 11, og stamceller 16. Konvensjonelt, er petriskåler og dekkglass brukes til å konstruere electrotactic kamre for å generere EFs 17. Slike enkle oppsett utgjøre problemer med middels fordampning og upresise EFs, men de kan overvinnes ved microfluidic enheter av lukkede, veldefinerte fluidic kanaler 12,18,19. Å systematisk studere cellulære responser i henhold til presise og kontrollerbare kjemiske og elektriske stimuli, vil det være av stor bruk for å utvikle microfluidic enheter i stand til å gi celler med flere stimuli samtidig. For eksempel Li et al. rapporterte en PDMS-baserte mikrofluidanordning for å lage én eller kjøl kjemiske gradienter og EFs 20. Kao et al. devrømte en lignende microfluidic chip å modulere kjemotaksis av lungekreftceller ved EFs 6. Dessuten, for å øke gjennomstrømningen, Hou et al. designet og fabrikkert en PMMA-baserte multikanal-dual-elektrisk felt chip for å gi celler med 8 forskjellige kombinerte stimuli, å være (2 EF styrker x 4 kjemiske konsentrasjoner) 21. For ytterligere å øke hele og legge skjærspenningen stimulans, utviklet vi to PMMA-baserte microfluidic enheter for å studere cellulære responser etter én eller kjøl kjemiske / elektriske / skjærspenning stimuli. Rapportert av Lo et al. 22,23, disse enhetene inneholder fem uavhengige cellekultur kanaler hvor det hele tiden fluidic flytende, etterligne in vivo sirkulasjonssystemet. I den første chip (kjemisk-skjærspenning chip eller CSS chip), fem relative konsentrasjoner på 0, 1/8, 1/2, 7/8 og 1 er generert i kultur regioner, og en skjærspenning gradient er prosert inne i hver av de fem dyrkningsområdene. I den andre brikken (kjemisk-elektriske felt chip eller CEF chip), ved hjelp av en enkelt sett av elektroder og 2 sprøytepumper, blir fem EF styrke generert i tillegg til fem forskjellige kjemiske konsentrasjoner innenfor disse dyrkningsområdene. designing microfluidic devices for studying cellular responses under single or coexisting chemical/electrical/shear stress stimuli

Microfluidic enhetene er i stand til å skape en nøyaktig og kontrollerbar cellulære mikromiljø pH, temperatur, saltkonsentrasjon, og andre fysiske eller kjemiske stimuli. De har vært vanlig benyttet for in vitro-cellestudier ved å tilveiebringe in vivo-som omgivelser. Spesielt hvordan cellene respons til kjemiske gradienter, elektriske felt, og skjærspenninger har trukket mange interesser siden disse fenomenene er viktig for å forstå mobilnettet egenskaper og funksjoner. Disse microfluidic brikker kan være laget av glass-substrater, silisiumskiver, polydimetylsiloksan (PDMS) polymerer, polymetylmetakrylat (PMMA) substrater eller polyetylentereftalat (PET) substrater. Ut av disse materialene, PMMA substrater er billig og lett kan behandles ved hjelp av laser ablasjon og skriving. Selv om noen få microfluidic enheter har blitt designet og fabrikkert for å generere flere, coexisting kjemiske og elektriske stimuli, ble ingen av dem anseseffektiv nok til å redusere eksperimentelle gjentar, spesielt for screening formål. I denne rapporten beskriver vi vår design og fabrikasjon av to PMMA-basert microfluidic chips for å undersøke cellulære responser, i produksjon av reaktive oksygenforbindelser og migrasjon under enkle eller coexisting kjemiske / elektriske / skjærspenning stimuli. Den første brikken genererer fem relative konsentrasjoner på 0, 1/8, 1/2, 7/8, og en i kultur regioner, sammen med en skjærspenning gradient produsert inne i hvert av disse områdene. Den andre brikken genererer de samme relative konsentrasjoner, men med fem forskjellige elektriske feltstyrker opprettet innenfor hver kultur område. Disse enhetene ikke bare gi celler med en presis, kontrollerbar mikro-miljø, men også øke den eksperimentelle gjennomstrømming.
