Chemosensation är ofta indelade i två sensoriska komponenter: lukt sinnen och smak sinnen. En viktig utmärkande egenskap hos dessa sinnen är att luktsinne (lukt) används för att ta prov på omgivande gasformiga kemiska miljön medan gustation (smak) kräver fysisk kontakt med en icke-flyktig substrat. Båda sensoriska modaliteter stimulera neurologiska svar som behandlas och avkodas i hjärnan att producera lämplig attraktiv eller frånstötande beteende 2. Dessa sinnen är därför avgörande för djurens överlevnad. Bananflugan Drosophila melanogaster är en modellorganism som fortsätter att växa i popularitet för användning i att förståing hur insekter uppfattar lukt och smak. Fruktflugor har enorma fördelar jämfört med andra modellsystem på grund av den rikedom av genetiska verktyg för dissektion av molekylära, cellulära och beteende vägar. Arbete under de senaste 15 åren har varit särskilt avgörande för att karakterisera de specifika cellulära identiteter, neuronala receptorer och signalering mekanismer som är involverade i både doft och smak. Nu, kraften i Drosophila-genetik används för att ytterligare klarlägga hur dessa processer är kodade vid den enda neuron och enkelkretsnivå 3-6. Därför analyser som ger ett enkelt mål avläsning av förändringar sensoriska vägar är avgörande för den fortsatta förväg av dessa områden. Vi beskriver här ett protokoll som kan användas för att fastställa smak preference i Drosophila. Vilken som helst kombination av dessa matkällor kan utnyttjas i denna experimentella utformning för att avgöra hur kända genetiska förändringar påverkar smak val. Olfactory and gustatory perception of the environment is vital for animal survival. The most obvious application of these chemosenses is to be able to distinguish good food sources from potentially dangerous food sources. Gustation requires physical contact with a chemical compound which is able to signal through taste receptors that are expressed on the surface of neurons. In insects, these gustatory neurons can be located across the animal's body allowing taste to play an important role in many different behaviors. Insects typically prefer compounds containing sugars, while compounds that are considered bitter tasting are avoided. Given the basic biological importance of taste, there is intense interest in understanding the molecular mechanisms underlying this sensory modality. We describe an adult Drosophila taste assay which reflects the preference of the animals for a given tastant compound. This assay may be applied to animals of any genetic background to examine the taste preference for a desired soluble compound.
