Som med alle andre modellen, kan den antigen-drevet kolitt modellen beskrevet ovenfor presentere noen problemstillinger som etterforsker utfører teknikken må være klar over. Dette antas å bestå av opptil 100 billioner mikrobielle celler og utgjør en av de mest tettbefolkede bakterielle habitater er kjent innen biologi 1-3. I GIT bakterier kolonisere en intestinal nisje hvor de overleve og formere 4. Til gjengjeld endows microbiota verten med flere funksjonelle funksjoner som ikke er kodet på sitt genom en. For eksempel microbiota stimulerer spredning av epitelceller, produserer vitaminer som er vert ikke kan produsere av seg selv, regulerer stoffskiftet og beskytter mot patogener 4-6. Gitt dette gunstige forhold, har noen forfattere foreslått at mennesker er &quot;super-organismer&quot; eller &quot;holobionts&quot; som er en blanding av bakterielle og menneskelige gener 7,8. Å opprettholde intestinal integritet krever et strengt regulert immun homeostase å holde balansen mellom toleranse og immunitet 13. En ubalanse i immun homeostase bidrar til induksjon av intestinale sykdommer slik som inflammatorisk tarmsykdom (IBD) 3,14. Det er to hovedtyper av IBD: Crohns sykdom (CD) og ulcerøs kolitt (UC). Pasienter med disse sykdommene vanligvis lider av rektal blødning, alvorlig diaré og magesmerter 15,16. Den eneste årsaken til IBD er fortsattukjent, men en kombinasjon av genetiske faktorer, miljøpåvirkninger og feilregulert immunresponser kan være nøkkelen begivenhet for sykdomsutvikling 15. Dyremodeller for IBD har vært brukt i over 50 år. I de siste tiårene nye IBD modellsystemer er utviklet for å teste ulike hypoteser om patogenesen av IBD 17,18. Den best karakteriseres modell for kronisk kolitt er den T-celle-transfer modell som induserer avbrudd av T-celle-homeostase 19,20. Denne modellen involverer overføring av naive T-celler fra immunkompetente mus inn i verter som mangler T- og B-celler (for eksempel RAG - / - og SCID-mus) 16,21. Utviklingen av sykdommen i denne modellen blir overvåket av 3-10 uker, ved å evaluere nærværet av diaré, redusert fysisk aktivitet, og tap av kroppsvekt. Dette er såkalt avmagringssyndromet 16. Sammenlignet med friske musene colonic vev av transplanterte verter er Thicker, kortere og tyngre 16. Ved hjelp av T-celle-overføring modell, er det mulig å forstå hvordan ulike T-cellepopulasjoner kan bidra til patogenesen av IBD 22. T-celleoverføring modell analyserer ikke interaksjoner mellom APC og T-celler i sykdomsprosessen i en antigen-spesifikk måte. Det har vist seg at en interaksjon mellom myeloide celler og lymfoide celler kan være ansvarlig for utviklingen av tarmbetennelse 23. Selv om mange aspekter av IBD er klarlagt, de første hendelsene som fører til sykdomsutviklingen fortsatt trenger å bli forstått. Det har vist seg at i fravær av bakterieflora overføring kolitt kan ikke bli etablert 24. Nylig har flere teorier foreslår at IBD kan være et resultat av en immunrespons mot kommensale bakterier 25. Forfatterne har også foreslått at commensal bakterier er viktig for å indusere betennelse i tarmen26. I bakterie frie (GF) dyr tarmimmunsystem er vanligvis svekket 27,28, men en kolonisering av disse mus med en blanding av spesifikt patogenfrie bakterier resulterer i utviklingen av fullt kompetente tarmimmunsystem 29. Derfor synes microbiota å være et sentralt element i patogenesen av IBD, enten som en mekanisme som predisponerer for eller beskytter mot utvikling av tarmbetennelse 30,31. Gjeldende teorier foreslår at IBD er et resultat av mikrobiell ubalanse, kalt dysbiosis, i genetisk disponerte pasienter 32, men det er ikke klart, men hvis dysbiosis er årsaken eller konsekvensen av sykdommen 12. Tatt i betraktning den rolle av mikroorganismer i utvikling av IBD, in vitro eksperimenter vist at CD4 + T-celler kan aktiveres ved APC pulsert med tarmbakterier 33,34. Videre har det blitt vist at antigener fraforskjellige commensal bakterielle arter, for eksempel E. coli, Bacteroides, Eubacterium og Proteus, er i stand til å aktivere CD4 + T-celler 35. Dette indikerer at presentasjonen av bakterielle antigener til T-celler er viktig for utvikling av IBD. Inflammatory bowel disease (IBD) is a chronic inflammation which affects the gastrointestinal tract (GIT). One of the best ways to study the immunological mechanisms involved during the disease is the T cell transfer model of colitis. In this model, immunodeficient mice (RAG-/- recipients) are reconstituted with naive CD4+ T cells from healthy wild type hosts.
This model allows examination of the earliest immunological events leading to disease and chronic inflammation, when the gut inflammation perpetuates but does not depend on a defined antigen. To study the potential role of antigen presenting cells (APCs) in the disease process, it is helpful to have an antigen-driven disease model, in which a defined commensal-derived antigen leads to colitis. An antigen driven-colitis model has hence been developed. In this model OT-II CD4+ T cells, that can recognize only specific epitopes in the OVA protein, are transferred into RAG-/- hosts challenged with CFP-OVA-expressing E. coli. This model allows the examination of interactions between APCs and T cells in the lamina propria.

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs).
