En outre, les souris transgéniques et knock-out peuvent être créés pour tester la fonction des produits spécifiques du gène dans un complexe physiologique ou de l&#39;environnement pathologique. De telles études imitent la pathologie humaine et peuvent fournir des informations mécaniste importantes liées à la découverte de nouvelles voies et de thérapies, ainsi que de fournir des détails importants pour caractériser les effets des médicaments sur la thrombose. Thrombus artériels pathologiques sont dues à une lésion endothéliale de couche ou d&#39;un dysfonctionnement et l&#39; exposition du flux sanguin vers la matrice subendothelial 3,4. Différents modèles de thrombose ont été développés pour induire ces dommages endothéliaux tels que les lésions mécaniques, une lésion par oxydation à base de Bengale composé photoréactif Rose et 5 blessure au laser. Dans ce spectre, le chlorure ferrique (FeCl 3) induite par une lésion vasculaire est un modèle largement utilisé de la thrombose. Ce réactif lorsqueappliquée sur la face externe des vaisseaux induit des dommages oxydatifs des cellules vasculaires 6-8, avec une perte de protection des cellules endotheliales à partir de plaquettes et les composants de la cascade de coagulation en circulation. Le modèle FeCl3 est simple et sensible à la fois des médicaments anticoagulants et antiplaquettaires, et a été réalisée sur la carotide et des artères fémorales, les veines jugulaires et mésentériques et les arterioles crémaster et veinules chez les souris, les rats, les cobayes et les lapins 6-15. Un paramètre mesurable dans ce modèle est le temps écoulé de la blessure à l&#39; occlusion complète du vaisseau, mesurée comme la cessation de l&#39; écoulement sanguin avec un débitmètre doppler ou sous observation directe avec la microscopie intravitale 6,7,9. Une plage de temps comprise entre 5 et 30 min a été rapporté dans plusieurs études chez des souris C57BL6 7-10,16, ce qui suggère que FeCl 3 concentrations, les types d&#39;anesthésie, les techniques chirurgicales, l&#39; âge de la souris, fond génomique, la méthode de mesure de bflux lood, et d&#39;autres variables environnementales ont des effets significatifs dans ce modèle. Cette grande variabilité rend difficile de comparer les études de différents groupes de recherche et peut détecter des différences subtiles difficiles. Une vision de minimiser ces variabilités et établir une manière uniforme et reproductible dans un système de modèle in vivo, nous avons affiné le modèle d&#39;artère carotide FeCl 3 induite par le fait que les données hautement reproductibles avec une variation minimale 6-10,16-19. Dans cet article, nous décrivons et partageons les compétences et présentons plusieurs exemples expérimentaux représentatifs qui peuvent bénéficier de ce modèle.

ferric chloride-induced murine thrombosis models

Arterial thrombosis (blood clot) is a common complication of many systemic diseases associated with chronic inflammation, including atherosclerosis, diabetes, obesity, cancer and chronic autoimmune rheumatologic disorders. Thrombi are the cause of most heart attacks, strokes and extremity loss, making thrombosis an extremely important public health problem. Since these thrombi stem from inappropriate platelet activation and subsequent coagulation, targeting these systems therapeutically has important clinical significance for developing safer treatments. Due to the complexities of the hemostatic system, in vitro experiments cannot replicate the blood-to-vessel wall interactions; therefore, in vivo studies are critical to understand pathological mechanisms of thrombus formation. To this end, various thrombosis models have been developed in mice. Among them, ferric chloride (FeCl3) induced vascular injury is a widely used model of occlusive thrombosis that reports platelet activation and aggregation in the context of an aseptic closed vascular system. This model is based on redox-induced endothelial cell injury, which is simple and sensitive to both anticoagulant and anti-platelets drugs. The time required for the development of a thrombus that occludes blood flow gives a quantitative measure of vascular injury, platelet activation and aggregation that is relevant to thrombotic diseases. We have significantly refined this FeCl3-induced vascular thrombosis model, which makes the data highly reproducible with minimal variation. Here we describe the model and present representative data from several experimental set-ups that demonstrate the utility of this model in thrombosis research.
