Les étapes critiques Il est essentiel de travailler dans des conditions d&#39;efficacité dans la fécondation in vitro. Ce protocole permet la manipulation des embryons de poisson zèbre dans la ploïdie, tels que la génération de diploïdes gynogénétiques homozygotes à partir haploïdes gynogénétiques (figure 1) ou la production de tétraploïdes. Ceci est réalisé en induisant un retard dans la cytocinèse correspondant exactement à un cycle cellulaire (figure 2A, 2B). Le retard touche un cycle cytokinèse est obtenu par le traitement par choc thermique. Le protocole standard de choc thermique (HS) tel que décrit à l&#39; origine par Streisinger et ses collègues impliqués une impulsion de température au cours de la période mpf 13-15, résultant en une un cycle de la division cellulaire décrochage au cours du premier cycle cellulaire 1. L&#39;efficacité de ce protocole a été récemment améliorée par balayage des cycles cellulaires début d&#39;une fenêtre temporelle de glissement du traitement par choc thermique. ploidy manipulation of zebrafish embryos with heat shock 2 treatment

Manipulation de ploïdie permet de transformations utiles, telles que les diploïdes à tétraploïdes ou haploïdes à diploïdes. Dans le Danio rerio zebrafish, en particulier la génération de diploïdes homozygotes gynogénétiques est utile dans l&#39; analyse génétique , car elle permet la production directe d&#39;homozygotes à partir d&#39; une seule mère hétérozygote. Cet article décrit un protocole modifié pour ploïdie duplication basée sur une impulsion de chaleur au cours du premier cycle cellulaire, Heat Shock 2 (HS2). Grâce à l&#39;inhibition de la duplication centriole, cette méthode se traduit par une division cellulaire précise décrochage au cours du deuxième cycle cellulaire. La précision d&#39;un cycle de division décrochage, couplé à pas affecté duplication d&#39;ADN, se traduit par la duplication du génome entier. Protocoles associés à cette méthode comprennent l&#39; oeuf et la collecte de sperme, traitement UV des spermatozoïdes, la fécondation in vitro et le pouls de la chaleur pour provoquer une division d&#39; une cellule de cycle retard et la duplication ploïdie. Une version modifiée de ce protocole peut être appliquépour induire des changements de ploïdie chez d&#39;autres espèces animales.

En dépit de l&#39;une des cellules cytokinèse du cycle de décrochage, la replication de l&#39; ADN se produit normalement dans ces embryons, ce qui entraîne la duplication du contenu d&#39;ADN de l&#39;embryon (figure 1).
