Detta protokoll möjliggör manipulering av ploiditeten i zebrafiskembryon, såsom i genereringen av homozygota gynogenetic diploider från gynogenetic haploider (Figur 1) eller produktion av tetraploids. Detta uppnås genom att framkalla en fördröjning av cytokines som motsvarar exakt en cellcykel (figur 2A, 2B). Nyckeln en-cykel fördröjning i cytokines uppnås genom behandling med värmechock. Standardprotokollet för värmechock (HS) såsom ursprungligen beskrivits av Streisinger och kollegor involverade en temperaturpuls under perioden 13-15 MPF, vilket resulterar i en en-cykel celldelning stall under den första cellcykeln 1. Effektiviteten av detta protokoll har nyligen förbättrats genom att scanna de tidiga cellcykler med en glidande tidsfönster värmechockbehandling. Detta scan identifierat en senare tidpunkt för en värmechock, fortfarande inom den första cellcykeln (22-24 MPF), som resulterar i en högre EMBRyos med en-cykel celldelning stall, som i detta fall påverkar den andra cellcykeln 2. Observationen att experimentella manipulationer under det första cellcykeln stör celldelningen under den andra cellcykeln och orsaka DNA-innehåll duplicering har även rapporterats i andra fiskarter 3,4. Vi hänvisar till denna modifierade protokoll som Heat Shock 2 (HS2 - termen &quot;2&quot; speglar att värmepulsen uppträder vid en senare tidpunkt än standard HS-metoden, och att cellcykelfördröjning orsakad av HS2 inträffar under den andra cellcykeln ). Dessa studier visade att grunden för cytokines rest efter värmechock är hämningen av centriol dubblering under värmepulsen, vilket påverkar spindelbildning och fåra induktion i följande cellcykeln. HS2 resulterar i utbyten av cellcykelstopp närmar 100%, och andelen ploiditeten dubbel upp till 4 gånger högre än standard HS 2. Embryon behandlade witha värmechock under blastomere cellcykel uppvisar många skadliga effekter, vilket tyder på att värmechock påverkar flera processer som krävs för celldelning 2. Å andra sidan, om värmechock appliceras före initieringen av cellcykel (tidsperiod 0-30 MPF), verkar det ha effekter av specifika störningar med centriol dubbelarbete och verkar inte påverka andra viktiga cellprocesser 2 . Dessa studier visade att den tid före inledningen av blastomere division verkar vara en utvecklande period mottaglig för att använda värmechock som ett verktyg för att specifikt manipulera ploiditet genom centriol inhibition. Den underliggande orsaken till den uppenbara selektivitet för värmechock på centriol dubblering är okänd, men kan relateras till en selektiv nedbrytning av centrosom substrukturer som observerats under värmepåfrestning i vissa celltyper, såsom leukocyter 5. Temporal synkronisering av embryonala deling åstadkommes genom in vitro-fertilisering (IVF). Användning av obehandlat spermier under gödslingsresulterar i diploida embryon som på HS2-inducerade en cykel cytokines stall blir tetraploid. Användning av UV-behandlade spermier, som bär tvärbindningar som inaktiverar dess DNA, resulterar i gynogenetic haploida embryon 6, som vid HSII-inducerade en cykel cytokines stall blir gynogenetic diploider 2. På grund av den resulterande hela genomet dubblering, de senare gynogenetic diploider är homozygot vid varje locus över genomet. För koncisa, hänvisar vi till gynogenetic haploida embryon som &quot;haploider&quot;, och homozygota gynogenetic diploida embryon som &quot;homozygota diploider&quot;. Om livskraftig och bördig, kan homozygota diploider användas för att initiera sterila och dödliga fria linjer. Direkt homozygoti inducerad av HS2 bör också lätt införlivas i genetisk analys eller genetiska skärmar, eftersom homozygota diploider från hondjur som är heterozygota bärare av mutheten uppvisar andelen homozygositet vid höga och fasta (50%) förhållanden 2. Följande protokoll beskriver åtgärder för att utföra HS2 och inducerar ploidi dubblering med full homozygoti. För tetraploid produktionen bör spermie lösning vara obehandlade. För homozygot diploid produktion, bör spermier inaktiveras genom UV-behandling. Zebrafisk mate främst under tre första hr om inledandet av deras ljus cykel 7, och både vuxna och ägg är känsliga för dygnsrytmen 8, så för bästa resultat IVF förfarande bör ske inom denna tidsperiod.

ploidy manipulation of zebrafish embryos with heat shock 2 treatment

Manipulering av ploiditet möjliggör användbara transformationer, såsom diploider till tetraploids eller haploider till diploider. I zebrafisk Danio rerio, generering av homozygota gynogenetic diploider är speciellt användbar i genetisk analys, eftersom det gör det möjligt att direkt framställning av homozygoter från en enda heterozygot mor.
