Hier beschrijven we een protocol om acuut letsel in de skeletspier (dat wil zeggen, de intramusculaire injectie van CTX) induceren. Zoogdieren volwassen skeletspieren worden gevormd door groepen van fascicules van meerkernige spiercellen (spiervezels), die gespecialiseerd zijn voor samentrekking zijn. Elke spiervezeloriėntaties een langwerpig syncytia, omringd door de sarcolemma (plasmamembraan) en die myofibrils, die bestaan ​​uit regelmatig en herhaaldelijk georganiseerde contractiele eiwitten (actine en myosine filamenten). Bovendien SC, inflammatoire en interstitiële cellen, angiogene processen en reïnnervatie gebeurtenissen betrokken stelt een gecoördineerde wijze repareren goed georganiseerde en specialized structuur. Er is grote belangstelling voor het bestuderen van verschillende aspecten van skeletspieren regeneratie niet alleen de fysiologie van de spier te begrijpen, maar ook voor therapeutische strategieën die diepere kennis over het gehele proces te verbeteren. Verschillende experimentele benaderingen zijn beschikbaar om de identiteit en functie van de verschillende celpopulaties, de signaalwegen en de moleculaire mechanismen te bestuderen. Muismodellen van acuut letsel vormen een krachtig hulpmiddel om vele aspecten van dit proces te onderzoeken. Andere gewoonlijk gebruikte technieken voor het induceren van acute spierschade kunnen onderzoekers het regeneratieproces volgen in vivo, van het prille begin tot het einde van het proces. Dit protocol beschrijft de stappen van de intramusculaire injectie van slangengif-afgeleide cardiotoxine (CTX), die myolyse induceert en activeert het regeneratieproces tot de analyse van weefselmonsters. Na CTX injectie, mice kunnen op verschillende tijdstippen opgeofferd afhankelijk experimentele eisen en de skeletspieren kan worden uitgesneden en verwerkt voor verdere analyse. Tenslotte beschrijven we de kleuring protocol van het weefsel secties om morfologische observaties en fundamentele kwantitatieve analyses uit te voeren. Dit protocol maakt het onderzoek naar acute skeletspier regeneratie in vivo op zeer reproduceerbare wijze 10.

Skeletspier aanpassing een fysiologisch proces dat plaatsvindt bij volwassen skeletspieren als reactie op letsel of ziekte. Acute verwonding geïnduceerde skeletspier aanpassing een veel gebruikt, krachtig modelsysteem om de gebeurtenissen betrokken bij spierregeneratie alsook de mechanismen en verschillende spelers bestuderen. Inderdaad, een gedetailleerde kennis van deze werkwijze is essentieel voor een beter begrip van de pathologische omstandigheden die leiden tot skeletspier degeneratie, en het helpt bij het identificeren van nieuwe gerichte therapeutische strategieën. De huidige werk beschrijft een gedetailleerd en reproduceerbare protocol voor acute skeletspier regeneratie in muizen te induceren via een enkele intramusculaire injectie van cardiotoxine (CTX). CTX behoort tot de familie van slangengif toxines en veroorzaakt myolyse van myovezels, die uiteindelijk leidt tot de regeneraties. De dynamiek van de skeletspieren regeneratie wordt geëvalueerd door histologische analyse van de spier secties. Het protocol ooktoont de experimentele procedures voor het ontleden, het invriezen en het snijden van de tibialis anterior spier, en de routine hematoxyline en eosine kleuring die op grote schaal wordt gebruikt voor latere morfologische en morfometrische analyse.

H &amp; E kleuring maakt de evaluatie van de morfologie van het regeneratieproces op specifieke tijdstippen tijdens skeletspier regeneratie.
