Un protocole similaire a été utilisée pour purifier le glutathion-S-transférase (GST) -tagged C terminaux 40 acides aminés de HCN1 (HCN1 c40) utilisé dans l&#39; étape 7 du protocole. A l&#39; étape 4, le facteur Z de l&#39;essai est calculée 21 pour une concentration donnée de TRIP8b (241-602) et HCN1 FITC pour garantir que l&#39;essai est approprié pour le criblage à haut débit (étape 5). Les étapes 6 et 7 sont des essais de validation pour confirmer que les résultats identifiés dans l&#39;écran à haut débit primaire agissent en perturbant l&#39;interaction entre TRIP8b (241-602) et HCN1 FITC plutôt que par un mécanisme non spécifique. Etape 7 utilise un plus grand HCN1 fragment C terminal (HCN1 C40) et emploie une analyse de proximité à base de billes, qui est basé sur le «tunnel» d&#39;un oxygène singulet à partir d&#39; un cordon de donneur à un bourrelet d&#39;accepteur amené à proximité les uns des autres par des protéines qui interagissent 22.

method for identifying small molecule inhibitors of the protein-protein interaction between hcn1 and trip8b

(HCN) canaux de nucleotide cyclique activé par hyperpolarisation sont exprimés de façon ubiquitaire dans le cerveau, où ils fonctionnent pour réguler l&#39;excitabilité des neurones. La distribution subcellulaire de ces canaux dans les neurones pyramidaux de la zone hippocampique CA1 est régulée par tétratricopeptide Rab8b contenant des protéines de répétition interagissant (TRIP8b), une sous-unité auxiliaire. Dans ce protocole, une description plus détaillée d&#39;un procédé de purification TRIP8b et l&#39;exécution d&#39;un écran à haut débit pour identifier les petites molécules inhibitrices de l&#39;interaction entre HCN et TRIP8b, est décrit. Le procédé de criblage à haut débit utilise une polarisation de fluorescence (FP) à base de test pour surveiller la liaison d&#39;un grand fragment de TRIP8b à un onze fluorophores marqués aminés peptide acide correspondant à la queue terminale HCN1 C. Cette méthode permet de composés «hit» d&#39;être identifiés sur la base de la variation de la polarisation de la lumière émise. tests de validation sont ensuite effectués pour veiller à ce que «hit» composés ne sont pas artifactual.

Pour éviter les problèmes de droits d&#39;auteur avec notre précédente publication, une sonde TAMRA-étiqueté HCN1 TAMRA a été utilisé pour générer des figures 2 et 3.
