Neste protocolo, uma descrição expandida de um método para purificar TRIP8b e a execução de uma tela de alto rendimento para identificar inibidores de pequenas moléculas da interacção entre o HCN e TRIP8b, é descrito. O método de rastreio de elevado rendimento utiliza uma polarização de fluorescência (FP) à base de ensaio para monitorar a ligação de um fragmento grande TRIP8b para um onze marcado com fluoróforo amino ácido péptido correspondente à cauda do terminal HCN1 C. Este método permite que compostos &quot;hit&quot; a ser identificado com base na alteração na polarização de luz emitida.
