Theiler&#39;s Murine Encefalomyelitis Virus (TMEV) een neurotrope enkelstrengs RNA virus dat voortdurend infecteert het muizen centrale zenuwstelsel (CNS). Bij gevoelige muizen, infectie met TMEV veroorzaakt een immuungemedieerde, chronische progressieve demyeliniserende ziekte, die bekend staat als TMEV-geïnduceerde diermodellen (TMEV-IDD). Experimentele infectie van muizen draait beloop lijkt op die waargenomen bij progressieve vorm van multiple sclerose (MS). TMEV-IDD wordt gekenmerkt door twee afzonderlijke fasen: de acute en de chronische fase. De acute fase een mild, gewoonlijk subklinische encefalitis 1,2. De tweede, de chronische fase begint ongeveer een maand na infectie bestaat uit een langzaam voortschrijdende beperking gekenmerkt door demyelinisatie, ontsteking en axonale beschadiging 1,2. De zwakte waargenomen bij muizen wordt geassocieerd met spasticiteit en soms ernstige tonic spasmen. Omdat er momenteel geen medications de progressieve handicap te verbeteren bij patiënten, onderzoekers in het bijzonder aangetrokken door TMEV-IDD, waardoor een optimale diermodel voor de bewaking van de impact van disease-modifying drugs op progressie van de ziekte vertegenwoordigt. In muizen en bij MS-patiënten, de controle van invaliditeitsprogressie vereist een continue klinische observatie gedurende langere tijdsperioden. Bij muizen, kan op lange termijn monitoring voor arbeidsongeschiktheid progressie worden bereikt met de Rotarod verrichtingsonderzoek. De Rotarod performance test is een standaard gedrags-test die door een motor betrokken functies, zoals coördinatie, balans, en vermoeidheid bij knaagdieren evalueert. De muizen moeten hun evenwicht op draaistang die draait onder continue versnelling te houden; de tijd latency te vallen van deze hengel wordt vastgelegd. Dieren met neurologische stoornissen zijn niet in staat om te verblijven op de roterende staaf zolang controles, en ze normaal afzetten wanneer de rotatiesnelheid hoger is dan hunmotorvermogen. Hoe meer neurologische stoornis de dieren, hoe eerder ze vallen uit de stang, en hoe korter de tijd latentie. Het voordeel van de Rotarod proef via traditionele visuele scoring systemen moet ernaar gestreefd genereert meten variabele-tijd-vertraging die uiteindelijk kan worden gebruikt voor statistische analyses om de effecten van therapieën en experimentele procedures 3 kwantificeren. In het Laboratorium voor Neuro-immunologie (LONI) in Dartmouth, zijn muizen blootgesteld aan een aanpassing protocol, waar ze worden getest voorafgaand aan TMEV infectie om hen vertrouwd te maken met de machine en hun normale &quot;basislijn&quot; balance coördinatie en motorische controle 4 te beoordelen, 5. Zodra de basislijn wordt vastgesteld en de muizen geïnfecteerd met TMEV, worden ze twee keer per week getest over een periode van enkele maanden. De werkelijke testprotocol duurt gemiddeld 150 dagen, waardoor een evaluatie vande daling van de balans, coördinatie en motorische controle over het gehele verloop van de demyeliniserende ziekte. Enkele honderden TMEV-IDD en sham-behandelde muizen werden tot nu toe getest op neurologische dysfunctie bij Dartmouth. Deze muizen hadden verschillende immunomodulerende behandelingen ontvingen, maar geen farmacologisch middel effectief is gebleken bij het verlichten invaliditeitsprogressie 6,7 te zijn. In dit artikel en de daarmee verband houdende protocol beschreven hoe u de progressieve neurologische stoornis die wordt weergegeven door TMEV-IDD muizen te karakteriseren. Vooral het protocol biedt aanbevelingen specifieke testparameters verondersteld algemeen geschikt voor het bestuderen van neurologische invaliditeit bij TMEV IDD-muizen met de Rotarod test. Deze procedure zorgt voor een basislijn waartegen om te beoordelen (1) de relevantie van deze muis model om progressieve MS en (2) het nut ervan voor het testen van therapieën die gericht zijn op de behandeling van progressieve neurologische aandoeningen zoals MS. Is duidelijk dat deRotarod prestatietest en actuele geoptimaliseerde testparameters en protocol zijn niet alleen nuttig bij het detecteren progressieve neurologische handicap in de TMEV-IDD muismodel, maar zijn ook bruikbaar bij het ontdekken stoornissen in andere virus-geïnduceerde en / of genetische muismodellen van neurodegeneratieve ziekten.

measuring progressive neurological disability in a mouse model of multiple sclerosis

After intracerebral infection with the Theiler's Murine Encephalomyelitis Virus (TMEV), susceptible SJL mice develop a chronic-progressive demyelinating disease, with clinical features similar to the progressive forms of multiple sclerosis (MS). The mice show progressive disability with loss of motor and sensory functions, which can be assessed with multiple apparatuses and protocols. Among them, the Rotarod performance test is a very common behavioral test, its advantage being that it provides objective measurements, but it is often used assuming that it is straightforward and simple. In contrast to visual scoring systems used in some models of MS, which are highly subjective, the Rotarod test generates an objective, measurable, continuous variable (i.e., length of time), allowing almost perfect inter-rater concordances. However, inter-laboratory reliability is only achieved if the various testing parameters are replicated. In this manuscript, recommendations of specific testing parameters, such as size, speed, and acceleration of the rod; amount of training given to the animals; and data processing, are presented for the Rotarod test.
