Aktivitetsbaseret proteinprofilering (ABPP) er en metode til identifikation af et enzym af interesse i en kompleks proteom ved anvendelse af en kemisk probe, som er målrettet mod enzymets aktive steder. En reporter tag indført i proben muliggør påvisning af det mærkede enzym ved in-gel fluorescens scanning, protein-blot, fluorescensmikroskopi eller væskekromatografi-massespektrometri. Her beskriver vi fremstilling og anvendelse af forbindelsen ARN14686, et klik kemi aktivitetsbaseret sonde (CC-ABP), som selektivt genkender enzymet N -acylethanolamine syre amidase (NAAA). NAAA er en cystein hydrolase, der fremmer inflammation ved at deaktivere endogene peroxisomproliferatoraktiveret receptor (PPAR) -alpha agonister, såsom palmitoylethanolamid (PEA) og oleoylethanolamide (OAS). NAAA syntetiseres som en inaktiv fuld længde proenzym, som aktiveres af autoproteolysis i det sure pH i lysosomer. Lokalisering undersøgelser have vist, at NAAA overvejende udtrykkes i makrofager og andre monocyt-afledte celler, såvel som i B-lymfocytter. Vi giver eksempler på, hvordan ARN14686 kan anvendes til at påvise og kvantificere aktiv NAAA ex vivo i gnavere væv ved protein-blot og fluorescensmikroskopi.

ARN14686 designet baseret på stilladset af NAAA inhibitor ARN726. Det 4-butyl-cyclohexylgruppe af ARN726 blev erstattet med en C9 mættet alifatisk kæde, der bærer en terminal alkyn tag (figur 1).
