Pancreatic β-Zellen sind die einzigen Insulin-produzierenden Zellen im Körper und sind kritisch Blutzucker Homöostase aufrechtzuerhalten. Während gesunde Individuen ausreichend β-Zellmasse aufweisen und Funktion richtig Blutzucker, Personen mit Diabetes durch unzureichende β-Zellmasse charakterisiert und / oder Funktions 1,2 regulieren. Es ist vorgeschlagen worden , dass β-Zell - Proliferation induzierende letztlich β-Zellmasse erhöhen und Glukose - Homöostase bei Patienten mit Diabetes 3 wieder herzustellen. Allerdings Evaluierung und Validierung von potentiellen β-Zell-proliferative Verbindungen in intakten Inseln ist notwendig, bevor wirksame Therapien entwickelt werden können. Die Entwicklung von Biomaterialien erhöht signifikant das Potential für eine gezielte Arzneimittelabgabe an eine Vielzahl von Zell- und Gewebetypen, einschließlich den pankreatischen β-Zellen. Hier beschreiben wir eine Methode zur Zusammenarbeit Kultur intakte Maus Inselchen mit biologisch abbaubaren Verbindung von Interesse (COI) beladene Poly (Milch-co-Glykolsäure) (PLGA) Mikrokugeln für die Zwecke der die Auswirkungen der anhaltenden Bewertung der in - situ - Freisetzung of mitogenen Faktoren auf β-Zellproliferation. Diese Technik beschreibt im Detail, wie PLGA-Mikrokügelchen zu erzeugen, um eine gewünschte Ladung unter Verwendung kommerziell erhältlicher Reagenzien enthält. Während die beschriebene Technik rekombinantes menschliches Bindegewebe-Wachstumsfaktor (rhCTGF) als Beispiel verwendet wird, könnte eine Vielzahl von COI ohne weiteres verwendet werden. Zusätzlich verwendet dieses Verfahren 96-Well-Platten, die Menge der Reagenzien zu minimieren, notwendig, β-Zellproliferation zu beurteilen.
