Formålet med næringspulsmetoden, som vist her, er for å karakterisere og kvantifisere responsen av den sterkt variert utvalg av omgivende strøm vann DOM over et dynamisk område av et tilsatt uorganisk næringsstoff. Oppløst organisk materiale (DOM) gir en viktig energi- og næringskilde til ferskvann økosystemer og er definert som organisk materiale som passerer gjennom en 0,7 mikrometer filter. Innenfor akvatiske økosystemer, kan DOM også påvirke lys demping og metallkompleks. DOM er en meget mangfoldig og heterogen blanding av organiske forbindelser med forskjellige funksjonelle grupper, så vel som essensielle næringsstoffer som nitrogen (N) og fosfor (P). Mens begrepet &quot;DOM&quot; beskriver hele bassenget inkludert dets C, N og P-komponenter, er dets konsentrasjon målt som oppløst organisk karbon (DOC). Den iboende molekylære kompleksitet DOM bassenget men skaper utfordringer for sin studie. For eksempel, er det ingen direkte måte å måle fraksjonen av den totale DOM pool bestående av organiske næringsstoffer slik som oppløst organisk nitrogen (DON) og oppløste organiske fosfor (DOP). I stedet må konsentrasjonen av organiske næringsstoffer bestemmes ved differansen ( <em&gt; f.eks [DON] = [totalt oppløst nitrogen] - [oppløst uorganisk nitrogen]). Ved å undersøke hvordan DOM bassenget svarer til et bredt spekter av eksperimentelt pålagt næringskonsentrasjoner vi håper å få bedre innsikt i hvordan DOM reagerer på varierende miljøforhold. En metode som vanligvis brukes i strømmen biogeokjemi er næringstilsetning metoden. Næringstilsetning eksperimenter har tradisjonelt blitt brukt for å forstå opptakskinetikk eller skjebnen av det tilsatte oppløste stoff 4,5,6,7. Nutrient tilleggene kan være kortsiktige på hr 6 til dag skalaen 4, eller langsiktige manipulasjoner i løpet av flere år 8. Nutrient tilleggene kan også omfatte isotopisk merket næringsstoffer (for eksempel 15 N-NO 3 -) for å spore lagt næringsstoffer gjennom biogeokjemiske reaksjoner. Men isotopen baserte studier er ofte erfansive og krever utfordrende analyser (f.eks digestions) av flere bunn avdelinger der isotopically merkede næringsstoffer kan beholdes. Siste eksperimentering har avdekket nytten av kortsiktige nærings pulser å belyse kontrollene ikke økende og omgivelses oppløsninger som DOM 9,10, avslører en ny måte ved å undersøke sanntid in situ biogeokjemiske reaksjoner. Her beskriver vi og demonstrere viktige metodiske skritt til å gjennomføre kortsiktige nærings pulser med sikte på å forstå kombinert biogeokjemi av C og N og spesielt kontrollene svært mangfoldig DOM bassenget. understanding dissolved organic matter biogeochemistry through in situ nutrient manipulations in stream ecosystems

Dissolved organic matter (DOM) is a highly diverse mixture of molecules providing one of the largest sources of energy and nutrients to stream ecosystems. Yet the in situ study of DOM is difficult as the molecular complexity of the DOM pool cannot be easily reproduced for experimental purposes. Nutrient additions to streams however, have been shown to repeatedly alter the in situ and ambient DOM pool. Here we demonstrate an easily replicable field-based method for manipulating the ambient pool of DOM at the ecosystem scale. During nutrient pulse experiments changes in the concentration of both dissolved organic carbon and dissolved organic nitrogen can be examined across a wide-range of nutrient concentrations. This method allows researchers to examine the controls on the DOM pool and make inferences regarding the role and function that certain fractions of the DOM pool play within ecosystems. We advocate the use of this method as a technique to help develop a deeper understanding of DOM biogeochemistry and how it interacts with nutrients. With further development this method may help elucidate the dynamics of DOM in other ecosystems.
