In vitro celle-telling er en viktig grunnleggende teknikk i et bredt spekter av vev kultur eksperimenter. Nøyaktig bestemmelse av antallet celler i en kultur er avgjørende for eksperimentell reproduserbarhet og standardisering 1,2. Celletelling kan utføres manuelt ved hjelp av en hemocytometer samt bruke en rekke automatiserte metoder, hver med sine egne fordeler og ulemper 3,4,5. De fleste av de automatiserte metoder for celletelling tilhører en av to klasser, de som bruker Coulter-prinsippet eller strømningscytometri. Coulter counter dra nytte av celler elektrisk motstand for å bestemme celleantallet og størrelse. De er raske, nøyaktige og billigere enn flowcytometere. For det første, ved farging av cellene med et fluorescerende fargestoff, dissosiasjon from membranen, og kvantifisering ved anvendelse av en fluorescerende leser 12. En begrensning ved denne fremgangsmåten for mengde er at ingen registrering kan holdes tilbake av membranene, og det er ingen mulighet for videre analyse 13. The National Institute of Healths ImageJ er et kraftig, fritt tilgjengelig bildebehandling suite. ImageJ har omfattende partikkel analyse algoritmer som kan brukes effektivt for å telle ulike biologiske partikler.
