Het belangrijkste doel van het huidige protocol is het volledig gescheiden neuronale en astrocytaire culturen, terwijl het handhaven ervan in gedeeld medium. Daarom moet de zuiverheid van de kweken verkregen wordt gecontroleerd bij het begin van de procedure. Gedurende de afgelopen decennia is de algemene interpretatie van neuroglia functie is ontstaan uit de toekenning van een louter ondersteunende naar een actieve regulerende rol met betrekking tot de neuronale functie 1. Vanwege hun prominente invloed op de hersenen homeostase in gezondheid en ziekte 2, astrocyten van bijzonder belang zijn voor de wetenschappelijke gemeenschap. In de afgelopen jaren diverse studies zijn gericht op neuron-glia interacties in vivo en in vitro 3. De meeste van de kweeksystemen niet voorziet in afzonderlijke analyse van beide celtypen en hun secretomen. Verschillende manieren gebruik maken van de directe cocultivatie van neuronen en glia om langdurige overleving en fysiologisch relevante neuronale ontwikkeling van het netwerk 4-6 te bereiken. Het huidige protocol dezelfde doelen bereikt, terwijl beide celtypen fysiek gescheiden 7. Vergeleken met conditioned medium benadert 8,9, ons systeem maakt het mogelijk om de bidirectionele communicatie tussen neuronen en astrocyten te bestuderen. the indirect neuron-astrocyte coculture assay: an in vitro set-up for the detailed investigation of neuron-glia interactions

Proper neuronale ontwikkeling en de functie is een voorwaarde van de ontwikkelingslanden en de volwassen hersenen. De mechanismen waardoor sterk gecontroleerde vorming en instandhouding van complexe neuronale netwerken niet volledig tot nu toe begrepen.
