Das Hauptziel des aktuellen Protokolls ist es, völlig getrennte neuronale und astrozytären Kulturen, während sie in gemeinsam genutzten Medium beibehalten wird. Aus diesem Grund erhalten die Reinheit der Kulturen sollten zu Beginn des Verfahrens überprüft werden. In den vergangenen Jahrzehnten hat sich die allgemeine Interpretation von neuroglia Funktion aus der Zuschreibung eines nur unterstützend auf eine aktive regulatorische Rolle in Bezug auf neuronale Funktion 1 entwickelt. Somit wurde gezeigt, dass die Zugabe an retinalen Ganglionzellen in vitro Thrombospondin die Bildung der Synapsen 13 induziert. Jedoch sind auch andere noch unbekannte Faktoren notwendig Synapsen zu machen funktionellen 13. the indirect neuron-astrocyte coculture assay: an in vitro set-up for the detailed investigation of neuron-glia interactions

Proper neuronaler Entwicklung und Funktion ist die Voraussetzung der Entwicklungs- und adulten Gehirn. Jedoch sind die Mechanismen, die die stark kontrollierten Bildung und Aufrechterhaltung von komplexen neuronalen Netzwerken zugrunde liegenden nicht vollständig bisher verstanden. Die offenen Fragen Neuronen in Gesundheit und Krankheit betreffen , sind vielfältig und das Erreichen von Verständnis der grundlegenden Entwicklung der menschlichen Pathologien zu untersuchen, zum Beispiel Alzheimer-Krankheit und Schizophrenie. Die detaillierte Analyse der Neuronen in vitro durchgeführt werden. Allerdings Neuronen fordern Zellen und brauchen die zusätzliche Unterstützung von Astrozyten für ihre langfristige Überleben. Diese zelluläre Heterogenität in Konflikt mit dem Ziel, die Analyse von Neuronen und Astrozyten zu sezieren. Wir stellen Ihnen hier eine Zellkultur-Assay, der für die langfristige Co-Kultivierung von reinen primären Neuronen und Astrozyten, die die gleiche chemisch definierten Medium gemeinsam nutzen können, während er physisch zu seingetrennt. In diesem Aufbau überleben die Kulturen für bis zu vier Wochen, und der Test ist für eine Vielzahl von Untersuchungen über Neuron-Glia-Interaktion geeignet.

Die Analyse der neuronalen Kulturen über den indirekten Cokultur-System ist vielfältig und kann in verschiedenen Stadien der Kultur Reifung durchgeführt werden. Aufgrund der Tatsache, dass die Zellen für bis zu 4 Wochen, sind Langzeituntersuchungen der Kulturen möglich aufrechterhalten werden kann.
