preparation of raav9 to overexpress or knockdown genes in mouse hearts

Контроль экспрессии или активности специфических генов через инфарктом доставки генетического материала в мышиных моделях позволяет исследовать функции генов. Их терапевтический потенциал в сердце может быть также определена. Существуют ограниченные подходы к молекулярной вмешательства в естественных условиях в сердце мыши. Рекомбинантный аденовирус-ассоциированный вирус (Раав) основе генной инженерии была использована в качестве основного инструмента для сердца манипуляции генов в естественных условиях. Особые преимущества этой технологии включают в себя высокую эффективность, высокую специфичность, низкий уровень геномную интеграции, минимальный иммуногенность, и минимальный патогенность. Здесь подробно процедура построения, упаковки и очистить векторы rAAV9 описано. Подкожное введение rAAV9 в неонатальных крысят приводит к надежной экспрессии или эффективной нокдауна гена (ов), представляющие интерес в сердце мыши, но не в печени и других тканях.
