Vi har visat en metod för att extrahera pigment från bläckfisk kromatofor granuler. Genom att särskilt rikta granulerna, är vårt mål att bestämma deras roll i att medla adaptiv färg. Denna metod skiljer sig från tidigare rapporter som syftar till att karakterisera bläckfiskpigment med hjälp av bulkvävnadsprover 14 eller frystorkad hud 15. Även om detta protokoll är effektiv på att utvinna kromatofor pigment, är det begränsat till små reaktions skalor....

Bläckfisk som bläckfisk, bläckfisk och bläckfisk har förmågan att dynamiskt ändra sitt utseende för att kamouflera och signalering. 1-6 Denna funktion stöds delvis genom selektiv areal utbyggnaden av pigmenterade organ kallas kromatofor. 4,7-9 kromatofor är mjuka ställdon som innehåller ett nätverk av nanostrukturerade pigmentkorn begränsade inom en cytoelastic säcken som är förankrad radiellt av muskelfibrer. 1,3 När de påverkas, kromatofor växa med 500% i presenterade yta fördela kornen hela organ. 3,7 , 10,11 När denna åtgärd samordnade över ett antal kromatofor den totala färgningen av djuret ändras. En matsmältning och homogeniseringsprocessen används sedan för att dissociera den omgivande vävnaden och separera kromatofor cellerna. De nanostrukturerade granuler isoleras därefter och renas från de återstående kromatofor som använder upprepad sonikering och centrifugering. Vid rening, pigment extraherade från granulerna i en process som är anpassad från utvinning av synlig färg från fjärilsvingar användning av sura metanollösningar. 13 Svepelektronmikroskopi (SEM) och spektrofotometri används för att bekräfta att de kromatofor pigmenten framgångsrikt extraheras med användning av denna process. Denna metod beskriver isolering av kromatofor granulat som används för att undersöka de molekylära bidrag till coloration i bläckfisk. 12 småmolekylära extraktioner från hela djur kan ofta vara en lång och mödosam process. Syftet är att informera framtida forskare i en effektiv och enkel protokoll för förvärvet av pigment från nanostrukturerade granulat i bläckfisk.

Cephalopods can undergo rapid and adaptive changes in dermal coloration for sensing, communication, defense, and reproduction purposes. These capabilities are supported in part by the areal expansion and retraction of pigmented organs known as chromatophores. While it is known that the chromatophores contain a tethered network of pigmented granules, their structure-function properties have not been fully detailed. We describe a method for isolating the nanostructured granules in squid Doryteuthis pealeii chromatophores and demonstrate how their associated pigments can be extracted in acidic solvents. To accomplish this, the chromatophore containing dermal layer is first manually isolated using a superficial dissection, and the pigment granules are removed using sonication, centrifugation, and washing cycles. Pigments confined within the purified granules are then extracted via acidic methanol solutions, leaving nanostructures with smaller diameters that are void of visible color. This extraction procedure produces a 58% yield of soluble pigments isolated from granules. Using this method, the composition of the chromatophore pigments can be determined and used to provide insight into the mechanism of adaptive coloration in cephalopods.
