Bakterielle biofilmer er grupper av mikroorganismer som er festet til hverandre og innkapslet i et selv utskilt matrise. 1,2 klassisk, representerer de bakterier fysisk festet til en abiotiske eller biotiske overflaten dannet under forhold med flyt. Biofilm har også vist seg å vokse i statiske betingelser (fravær av strømning) og distal fra overflater, slik som ved luft-væske-grensesnittet av termiske bassenger eller pellicles dannet i reagensglass. Disse biofilm har lenge vært anerkjent i miljøet og er en stor ulempe for industrielle prosesser, som de kan dannes i vannreservoarer eller i rør, noe som resulterer i begroing, korrosjon og blokkeringer. 3,4 Biofilmer er også avgjørende i helsevesenet, ettersom de har vist seg være involvert i kateterrelaterte infeksjoner, lungeinfeksjoner hos pasienter med cystisk fibrose, så vel som i en rekke andre infeksjoner. 5,6 Ett av kjennetegnene på biofilm infeksjoner er deøkt mottakelighet av bakterier mot antibiotika og nedsatt clearance av det medfødte immunsystemet. 7-9 Den mest studerte, klinisk relevante scenarier med biofilm-basert infeksjon oppstår hos pasienter med cystisk fibrose (CF), som er kronisk infisert med Pseudomonas aeruginosa biofilm. P. aeruginosa kan gjennomgå en rekke endringer i løpet av opprettelsen av kronisk infeksjon som gjør det svært vanskelig å behandle. 10,11 Biofilm kan forskjellig aktivere medfødt immunitet og kjøre betennelse. 12-14 Ettersom disse infeksjoner føre til økt sykelighet og dødelighet hos CF-pasienter, er det viktig å forstå faktorer som kan påvirke biofilm utvikling i denne sammenheng. Biofilm består av grupper av bakterier innkapslet i en selv utskilt matrise. De spiller en viktig rolle i industriell forurensning samt i utvikling og utholdenhet av mange helserelaterte infeksjoner. En av de mest godt beskrevet og undersøkt biofilm i human sykdom oppstår ved kronisk pulmonar infeksjon i cystisk fibrose pasienter. Når man studerer biofilm i sammenheng med verten, kan mange faktorer påvirker biofilmdannelse og utvikling. For å identifisere hvordan vertsfaktorer kan påvirke biofilmdannelse og utvikling, ble det benyttet en statisk kammer, dekkglass metode for å dyrke biofilmer i nærvær av verts-avledede faktorer i form av sputum-supernatanter. Bakterier blir sådd inn i kammer og utsatt for sputum filtrater. Etter 48 timer med vekst, er biofilm farget med et kommersielt biofilm levedyktighet kit før konfokal mikroskopi og analyse. Etter bilde oppkjøpet, kan biofilm egenskaper vurderes ved hjelp av ulike programvareplattformer.Denne metoden tillater oss å visualisere viktige egenskaper biofilm vekst i nærvær av forskjellige stoffer, inkludert antibiotika.

Den generelle utformingen av forsøket er vist i figur 1.
