Bacteriën kunnen schakelen tussen twee zeer verschillende levensstijlen, plankton en zittend, waarvan een biofilm is de meest voorkomende voorbeeld. In biofilms, bacteriën vormen gestructureerde gemeenschappen ingebed in een zelfgeproduceerde matrix 1. Dit zelf geproduceerde matrix is ​​een barrière tussen de bacteriën en hun externe omgeving, en het beschermt de microbiële cellen, waardoor ze in de buurt. De enige bestaande gestandaardiseerde screening-toepassing test is de zogenaamde Calgary Biofilm apparaat, waaruit de handel verkrijgbare minimale biofilm elimineren concentratie (MBEC) systeem ontwikkeld 13-15. De beperking van deze test is dat de biofilms worden geteeld op pennen, en niet elke bacteriële species of stammen binnen dezelfde soort kunnen biofilms deze inrichting te vormen. Bovendien methoden die met name kunnen worden toegepast op de exploratie van natuurlijke verbindingennodig. Natuurlijke producten zijn de belangrijkste bron voor innovatie in antimicrobiële drug discovery in de afgelopen eeuw 16 geweest. Ze kunnen nieuwe anti-biofilm verbindingen te voorzien van unieke werkingsmechanismen die ook effectief tegen persistorcellen kan zijn. Zo is de exploratie van natuurlijke en natuurlijk geïnspireerd bibliotheken heeft een hoge kans op het produceren van veelbelovende en unieke anti-biofilm leads. Hier presenteren we de experimentele gegevens van een platform van assays die is ontwikkeld voor het screenen van chemische anti-biofilm verbindingen met behulp van drie tests om de effecten op de levensvatbaarheid, totale biomassa en matrix van Staphylococcus aureus biofilms meten. De eerste assay meet biofilm levensvatbaarheid en het is gebaseerd op resazurin kleuring. Resazurin is een redox vlek die is blauw en niet-fluorescerende in zijn geoxideerde toestand en kleurt roze, sterk fluorescerende resorufine wanneer het gereduceerd door metabolische activiteit van de bacteriën. Het is een zeer eenvoudige en snelle methode geschikt voor primaire screenings 17-20. De tweede test, op basis van kristalviolet kleuring, meet totale biofilm massa. Crystal violet is een veel gebruikte vlek voor het bestuderen van bacteriën en bacteriën in biofilms 19,21-23. De test is gebaseerd op goedkope reagentia en heeft een eenvoudige absorptie eindpunt lezen. Het derde onderzoek richt de extracellulaire polymere stoffen (EPS) -matrix van de biofilm via tarwekiem agglutinine (WGA), dat specifiek bindt aan N-acetyl-glucosamine residuen (PNAG) aanwezig in de matrix op Staphylococcus biofilms 24 poly-. De WGA is geconjugeerd met een fluorofoor die kan worden gedetecteerd via fluorescentie-intensiteit lezers 25. Wij presenteren hier de reden en de details van het platform dat we ontwikkeld, met inbegrip van voorbeelden van toepassingen.

a platform of anti-biofilm assays suited to the exploration of natural compound libraries

Biofilms worden beschouwd als een van de meest uitdagende onderwerpen moderne biogeneeskunde en het potentieel verantwoordelijk voor meer dan 80% van antibiotica-tolerante infecties. Biofilms hebben een uitzonderlijk hoge tolerantie voor chemotherapie, waarvan wordt gedacht multifactoriële te zijn weergegeven. Bijvoorbeeld, de matrix levert een fysieke barrière die het binnendringen van antibiotica vermindert in de biofilm. Ook cellen in de biofilm fenotypisch verschillend. Deze situatie heeft een progressieve paradigmaverschuiving in drug discovery, waarin op zoek naar anti-biofilms werd aangespoord om een ​​meer prominente plaats innemen gereden. Een bijkomend probleem is dat er eenzeer beperkt aantal gestandaardiseerde biofilm onderzoek, met name die kan worden gebruikt voor grote verwerkingscapaciteit screenen van chemische bibliotheken. Hier wordt een experimentele anti-biofilm platform voor chemische screening gepresenteerd. Het maakt gebruik van drie bepalingen om biofilm levensvatbaarheid (met Resazurin kleuring), totale biomassa (met kristalviolet kleuring) en biofilm matrix (gemeten met behulp van een tarwekiem agglutinine, WGA-fluorescentie-gebaseerde kleuring van het poly-N-acetyl-glucosamine, PNAG , fractie). Alle testen werden ontwikkeld met behulp van Staphylococcus aureus bacteriën als model. Voorbeelden van het platform kan worden gebruikt voor primaire screening en voor functionele karakterisering van geïdentificeerde anti-biofilm hits worden gepresenteerd. Deze experimentele sequentie verder zorgt voor de indeling van de treffers op basis van de gemeten eindpunten. Het biedt ook informatie over hun werkingsmechanisme, vooral op lange termijn versus korte termijn chemotherapeutische effecten. Het is dus zeer Advantageous voor de snelle identificatie van hoogwaardige hit verbindingen die als uitgangspunt kunnen dienen voor diverse biomedische toepassingen.

In het voorgestelde platform, worden de effecten op de levensvatbaarheid, biomassa en biofilm matrix gekwantificeerd.
