Inflammation är iscensatt av en uppsjö av cytokiner och kemokiner, vilka i sin tur har förmåga att attrahera andra immunceller till platsen för infektion och för att inducera kardinal tecken på inflammation, såsom rodnad, svullnad, smärta, förlust av funktion, och feber . Varaktighet och intensitet av inflammation beror på flera faktorer, men lösa inflammation och återställa homeostas är ett kritiskt steg för att undvika uppkomsten av kroniska inflammatoriska sjukdomar. Nya framsteg inom neurovetenskap och immunologi har unraveled specifika neurala mekanismer med enorm terapeutisk potential för att styra inflammation både i det centrala nervsystemet och i periferin. En av dessa mekanismer är den kolinerga antiinflammatoriska vägen (CAP), även känd som den inflammatoriska reflex, som drivs av det autonoma nervsystemet 4, 5. Det är för närvarande tänkt att inflammatoriska mediatorer aktiverar sensoriska nerver och skicka signaler om tillståndet av inflammation till det centrala nervsystemet. En reflexsvar aktiveras sedan genom den efferenta vagusnerven. En omfattande studie om de anatomiska detaljerna i CAP har avslöjat en parasympatiska-sympatisk modell består av två nerver, vagusnerven och mjälten nerv, respektive 6. Medan vikten av nikotinreceptorer (α7nAChR) i mjälten har påvisats tidigare 9, muskarinreceptorer (mAChR) är obligatoriska för den centrala aktivering av reaktionsvägen 10, 11. Mer nyligen, perifer administrering av en centralt verkande M1 muskarinagonist signifikant undertryckta i serum och mjälte tumörnekrosfaktor α (TNFa) under letal murin endotoxemi, en åtgärd som krävs intakt vagusnerv och mjält nerv signalering 12. Vi har också visat nyligen att möss som saknar prostaglandin E2 (PGE2) kunde inte svara på vagusnerven stimulering och inte nedreglera LPS-inducerad frisättning av cytokiner i serum och mjälte 3. Därför kan den gemensamma jordbrukspolitiken också regleras av andra än huvud ACh pathw systemay. Vagusnerven har utsetts som sådan på grund av dess vandrande kurs i kroppen, innervating huvudorgan, inklusive lever, lungor, mjälte, njurar och tarm 13. Med tanke på denna stora innervation och mycket potent immunsuppressiv effekt av vagusnerven kan den terapeutiska potentialen hos den gemensamma jordbrukspolitiken omfattar ett brett spektrum av inflammatoriska tillstånd. Vagusnerven kan vara elektriskt (eller mekaniskt) aktiveras, med kontroll över spänning och frekvens, och i motsats till konventionell behandling, utan läkemedel läggas till kroppen. Prövningar pågår i reumatiska patienter, till exempel för att testa den kliniska betydelsen av VNS för behandling av kronisk inflammation 14. Helt och hållet, den neuro-immuna kommunikation och reglering av inflammation är för närvarande under utredning, vilket kommer att ge ett möjligt alternativ behandling till konventionell terapi. Därför, analys av vagusnerven stimuleringn effekt i de olika innerverade organ, men även karakterisering av potentiella terapeutiska verkan i djurmodeller av kronisk inflammation, skulle definitivt ge insikter och hoppas på nya potentiella terapeutiska mål. Den ursprungliga metod som utvecklats av Tracey och kollegor 4 kunde inte överföras till vårt forskningsområde på grund av överstimulering av det inflammatoriska svaret (med en dödlig dos av LPS) och en alltför kort tidsintervall mellan CAP aktivering och avläsning. Inflammation is a local response to infection and tissue damage mediated by activated macrophages, monocytes, and other immune cells that release cytokines and other mediators of inflammation. For a long time, humoral and cellular mechanisms have been studied for their role in regulating the immune response, but recent advances in the field of immunology and neuroscience have also unraveled specific neural mechanisms with interesting therapeutic potential. The so-called cholinergic anti-inflammatory pathway (CAP) has been described to control innate immune responses and inflammation in a very potent manner. In the early 2000s, Tracey and collaborators developed a technique that stimulates the vagus nerve and mimics the effect of the pathway. The methodology is based on the electrical stimulation of the vagus nerve at low voltage and frequency, in order to avoid any side effects of overstimulation, such as deregulation of heart rate variability. Electrical devices for stimulation are now available, making it easy to set up the methodology in the laboratory. The goal of this research was to investigate the potential involvement of prostaglandins in the CAP. Unfortunately, based on earlier attempts, we failed to use the original protocol, as the induced inflammatory response either was too high or was not suitable for enzymatic metabolism properties. The different settings of the original surgery protocol remained mostly unchanged, but the conditions regarding inflammatory induction and the time point before sacrifice were improved to fit our purposes (i.e., to investigate the involvement of the CAP in more limited inflammatory responses). 
The modified version of the original protocol, presented here, includes a longer time range between vagus nerve stimulation and analysis, which is associated with a lower induction of inflammatory responses. Additionally, while decreasing the level of lipopolysaccharides (LPS) to inject, we also came across new observations regarding mechanistic properties in the spleen.
