EA ist eine alternative Medizin, die therapeutische Wirkung bei Epilepsie aufweisen kann und hat in der alten chinesischen Literatur dokumentiert. Epilepsie ist eine häufige neurologische Erkrankung, bei der wiederkehrenden Anfällen während eines Patienten Lebensdauer auftreten. Die meisten epileptischen Rezidive können werden gut kontrollierte durch Antiepileptika (AEDs). Doch etwa 30% der Patienten mit Epilepsie entwickeln refraktärer Epilepsie 1. Epilepsie Ursachen Schlafstörungen, die weitere Wiederholung verschlimmern können. Der Nachweis zeigt , dass Epilepsie entweder Schlaf in der Nacht stören können oder übermäßige Tagesschläfrigkeit 2,3 verursachen kann. Unsere bisherigen Untersuchungen zeigen weiter , dass Epilepsie bei Zeitgeber Zeit auftritt (ZT) 0, dh der Beginn der Lichtperiode im Licht: Dunkel - Zyklus nimmt Schlaf; dies wird durch Corticotropin-Releasing-Hormon (CRH), einen homöostatischen Faktor vermittelt. Epilepsie bei ZT13 (der Beginn der Dunkelperiode) erhöht die Expression eines anderen homöostatischen Faktor, Interleukin-1 (IL-1), die Schlaf erhöht. Schlaf-Tagesrhythmus verändert werden, wenn Epilepsie bei ZT6 auftritt, der Mitte desLichtperiode 4,5. Auf der anderen Seite, Schlafstörungen weiter die Progression und ein erneutes Auftreten von 6 Epilepsie verschlimmern. Auf der Grundlage der oben genannten Beweise, versuchen wir eine optimale therapeutische Methode, um gleichzeitig zu offenbaren Epilepsie kontrollieren und Schlaf Störungen bei Epilepsie-Patienten zu verhindern. Wir fanden zuvor , daß electroacupuncture (EA) mit einer 10-Hz Stimulationsfrequenz, bei dem eine bestimmte Menge an Strom in die acupoint durch eine Edelstahladel geliefert wird, erfolgreich unterdrückt Elektrokortikogramm (ECoG) epileptische Aktivitäten und Epilepsie-induzierter Schlafstörungen 7 . EA mit einer Stimulationsfrequenz von 100 Hz weiter verschlechtert epileptischen Aktivitäten und Schlaf Störungen bei Ratten 8,9. Dieses erfolgreiche Experiment hängt von drei Faktoren ab: erstens, eine machbare epileptischen Tiermodell; zweitens ein Verfahren zur Schlaf Aufzeichnung und Analyse in Nagetieren; und drittens die genaue Leistung der Akupunktur und die Genauigkeit der acupoint locations. Epilepsie ist in zwei Haupttypen unterteilt: fokalen Epilepsie und generalisierter Epilepsie. Wir interessieren uns für fokale Temporallappen-Epilepsie (TLE), generalisierter Epilepsie, Status epilepticus (SE) und die Wiederholung der spontanen generalisierte Epilepsie. Daher werden verschiedene Manipulationen angewendet geeigneten epileptische Modelle für unsere Experimente erstellen. Herzustellen Brenn TLE, eine niedrige Dosis von Pilocarpin wird in den linken zentralen Kern der Amygdala (CEA) verabreicht. Um dieses Modell zu überprüfen, sechs ECoG Elektroden auf der frontalen (F1 &amp; F2), parietalen (P1 &amp; P2) implantiert und okzipitalen (O1 &amp; O2) in den beiden Lappen der linken und rechten Hemisphären, und weitere zwei Referenzelektroden (R1 &amp; R2) über die Kleinhirns in beiden Hemisphären angeordnet. Eine zusätzliche Mikroinjektionsführungskanüle wird operativ in die linke CeA implantiert (AP, 2,8 mm von Bregma; ML, 4,2 mm; DV, 7,8 mm, bezogen auf Bregma). Die Koordinaten werden von der Paxinos und W angepaßtATSON Ratte Atlas 10. Wenn die Brenn TLE erfolgreich induziert wird, wird nur die Aufzeichnung von der Elektrode auf der linken parietalen Kortex (P1), die in der Nähe der linken CeA ist, sollte die dominant epileptiforme ECoGs, ohne signifikante epileptiforme ECoGs erwerben von den anderen ECoG Elektroden aufgezeichnet. Intraperitoneale (IP) Injektionen von Pilocarpin in Ratten induzieren Epilepsie und SE verallgemeinert, aber dies kann tödlich sein. Fünf IP-Injektionen von pentylenetetrazol (PTZ) mit einem Ein-Tages-Intervall zwischen jeder Injektion erfolgreich spontane generalisierte Epilepsie bei Mäusen induzieren und auch dafür sorgen, das Überleben der Mäuse. Zwei Draht ECoG Elektroden werden in die frontalen und parietalen Kortex in den Mäusen implantiert erhalten ECoG Signale und spontan wiederkehrende Epilepsie zu überprüfen. Polysomnographie (PSG) ist eine umfassende Methode physiologischen Veränderungen aufzuzeichnen, die während des Schlafes auftreten, und es kann objektiv Schlaf in verschiedene Stadien der nicht-schnelle Augenbewegung (NREM) und r klassifizierenapid Augenbewegung (REM) Schlaf. PSG zeichnet Parameter von Körperfunktionen, einschließlich der Gehirnwellen (Elektroenzephalogramm, EEG), Augenbewegungen (Elektrookulogramm, EOG), Skelettmuskel Töne (Elektromyogramm, EMG), Herzrhythmus (Elektrokardiogramm, EKG) und Sauerstoffgehalt im Blut und Atmungsparameter. Bei Ratten, wir ECoGs, EMGs, Rindentemperatur und Bewegungsaktivität aufzuzeichnen Wachsamkeit Zustände in Wachheit, NREM Schlaf und REM-Schlaf zu klassifizieren. Schlaf-Analyse bei Mäusen ist ECoGs, EMGs und Bewegungsaktivität Ergebnisse mit durchgeführt. Die Ratten werden chirurgisch mit drei ECoG Schraube Elektroden an der Stirn-, Scheitel- und kontralateralen Kleinhirns Rinden von stereotaktischen Operation implantiert. Nach der Akquisition Bestimmung der Wachsamkeit Staaten (Wachheit, NREM Schlaf und REM-Schlaf) durchgeführt wird entsprechend den Parametern aus den ECoGs erworben, EMG, Gehirntemperatur und der Bewegungsaktivität. Detaillierte Kriterien für das Verhalten des Tieres zu kategorisieren beiden bei Ratten und Mäuse werden in t beschriebener Protokoll. Ratten und Mäuse müssen sich mit einer niedrigen Dosis von zoletil (25 mg / kg) anästhesiert werden, die normalerweise während der stereotaktischen Chirurgie verabreicht Hälfte der Dosis von Anästhetika ist, vor der manuellen Akupunktur oder EA durchführt. Diese Dosierung erlaubt Tiere 30 bis 35 Minuten nach der Injektion zu wecken. Entweder manuell Akupunktur oder EA wird zu Beginn der Dunkelperiode durchgeführt wird, mit einer konstanten Zeitdauer von 30 min, und jedes Tier nacheinander für zwei bis drei Tage behandelt. Stimulierender EA Ströme werden in einen bestimmten acupoint durch eine Edelstahl Nadel geliefert, die in die acupoint eingesetzt ist. Der Reizstrom ein Zug von zweiphasigen Rechteckimpulse ist, bei dem die Impulsdauer beträgt 150 ms und die Stimulationsintensität von 1 mA. Der schwierige Teil der manuellen Akupunktur oder EA ist die Akupunkturpunkte bei Nagetieren zu lokalisieren. Die location von acupoints in Ratten oder Mäuse ähnelt in ihrer anatomischen Lage beim Menschen. Zum Beispiel werden die bilateralen Fengchi Akupunkte 3 mm entfernt von der hinteren Mittellinie an dem Hals zwischen den beiden Ohren befinden, die zu seiner anatomischen Lage beim Menschen 11 ähnlich ist. Darüber hinaus können die Akupunkturpunkte mit niedriger Impedanz auf der Haut weiter bestätigt werden. Sham-Akupunktur oder Schein-EA Manipulation ist für Akupunktur oder EA Experimente notwendig. Sham - Akupunktur oder Schein - EA sollte in der Nähe der acupoint, wie in der Nähe der Achselhöhle 12 befindet sich in einem nicht acupoint durchgeführt werden. Ancient Chinese literature has documented that acupuncture possesses efficient therapeutic effects on epilepsy and insomnia. There is, however, little research to reveal the possible mechanisms behind these effects. To investigate the effect of acupuncture on epilepsy and sleep, several issues need to be addressed. The first is to identify the acupoints, which correspond between humans, rats, and mice. Furthermore, the depth of insertion of the acupuncture needle, the degree of needle twist in manual needle acupuncture, and the stimulation parameters for electroacupuncture (EA) need to be determined. To evaluate the effects of acupuncture on epilepsy and sleep, a feasible model of epilepsy in rodents is required. We administer pilocarpine into the left central nucleus of the amygdala (CeA) to simulate focal temporal lobe epilepsy (TLE) in rats. Intraperitoneal (IP) injection of pilocarpine induces generalized epilepsy and status epilepticus (SE) in rats. Five IP injections of pentylenetetrazol (PTZ) with a one-day interval between each injection successfully induces spontaneous generalized epilepsy in mice. Recordings of electrocorticograms (ECoGs), electromyograms (EMGs), brain temperature, and locomotor activity are used for sleep analysis in rats, while ECoGs, EMGs, and locomotor activity are employed for sleep analysis in mice. ECoG electrodes are implanted into the frontal, parietal, and contralateral occipital cortices, and a thermistor is implanted above the cerebral cortex by stereotactic surgery. EMG electrodes are implanted into the neck muscles, and an infrared detector determines locomotor activity. The criteria for categorizing vigilance stages, including wakefulness, rapid eye movement (REM) sleep, and non-REM (NREM) sleep are based on information from ECoGs, EMGs, brain temperature, and locomotor activity.
