Het doel van de onderhavige protocol was een hoge resolutie respirometrie om mitochondriale ademhalingsketen complexen (I-IV) ademhalingssnelheid, maximale mitochondriale elektronentransport systeemcapaciteit en buitenste mitochondriale membraan integriteit te meten. Er zijn een aantal kritische stappen in het huidige protocol. Eerst worden cellen zuurstofverbruik meestal genormaliseerd naar het aantal cellen (pmol / [sec x aantal cellen]). Mitochondriën vervullen een belangrijke rol in de cellulaire energiemetabolisme, vooral door toepassing van zuurstof aan adenosine trifosfaat (ATP) te produceren. Ze zijn betrokken bij celdood en in verschillende humane ziekten. Mitochondriale oxidatieve fosforylering (OXPHOS) combineert elektron transport langs de elektronen transportketen met zuurstofverbruik en ATP synthese. De mitochondriale tricarbonzuur (TCA) cyclus is betrokken bij de omzetting van eiwitten, koolhydraten en vetten in energierijke verbindingen nicotinamide adenine dinucleotide (NADH) en flavineadeninedinucleotide (FADH 2). Elektronen van NADH en FADH 2 worden vervolgens overgebracht naar de luchtwegen elektronentransportketen eiwitcomplexen (I tot IV) in het binnenste mitochondriale membraan. Bovendien kunnen twee andere redox pathways elektronen overdragen aan elektronen transportketen: i) mitochondriale elektron overbrengende flavoproteïne (ETF) dat zich op het vlak van de matrix binnenste mitochondrial membraan en levert elektronen van vetzuur β-oxidatie; en ii) mitochondriale glycerofosfaatdehydrogenase die glycerofosfaat oxideert tot dihydroxyacetonfosfaat en voedt elektronen de mitochondriale elektronentransportketen. Complex IV (de uiteindelijke elektronenacceptor) kan de elektronen een zuurstofmolecuul omzetten zuurstof twee moleculen water. Verplaatsen van de elektronen van respiratoire elektronentransportketen complex I tot IV wordt gekoppeld proton stroom vanaf de mitochondriale matrix intermembrane de ruimte die een elektrochemische gradiënt over de mitochondriale binnenmembraan vaststelt. Daarna mitochondriale complex V (ATP synthase) shuttles waterstofionen terug in de mitochondriale matrix en synthetiseert ATP moleculen. OXPHOS functie in vivo en in vitro gebruik van verschillende technieken en verschillende mitochondriale respiratie toestanden kunnen worden verkregen worden beoordeeld. In geïsoleerde mitochondria de volgende respiratory toestanden kunnen worden gemeten: i) basaal mitochondriale respiratie (stand 1), ii) zuurstofverbruik na toevoeging van specifieke substraten van de mitochondriale ademhalingsketen complexen (stand 2), iii) maximale mitochondriale zuurstofconsumptie na toevoeging van verzadigende concentraties adenosine difosfaat (ADP) (toestand 3) en, iv) rusten ademhaling na ADP verbruik (omgerekend naar ATP) (staat 4). In intacte cellen kan de volgende luchtwegen toestanden worden gemeten: i) basale cellulaire zuurstofverbruik in aanwezigheid van endogene substraten en ADP, ii) basale cellulaire zuurstofverbruik in aanwezigheid van oligomycin (oligomycin ongevoelige ademhaling) en oligomycine-gevoelige ademhaling (ATP omzet), iii) FCCP ontkoppeld ademhaling, en iv) niet-mitochondriale respiratie na toevoeging van antimycin A en rotenon. In gepermeabiliseerde cellen kunnen specifieke substraten van de elektronentransport keten complexen en ADP toegevoegd en maximale complexe afhankelijke ademhalingssnelheid such zo complex I, II en IV-afhankelijke respiratoire tarieven kunnen worden gemeten. Metingen van cellulaire ademhaling belangrijke inzichten in mitochondriale ademhalingscapaciteit specifieke complexen I-IV, mitochondriale integriteit en energiemetabolisme 1, 2, 3. Een van de hulpmiddelen die metingen van mitochondriale zuurstofverbruik met een hoge nauwkeurigheid, resolutie en gevoeligheid in staat is de hoge-resolutie oxygraph 4. De hoge resolutie oxygraph apparaat bevat twee kamers met injectiepoorten en elke kamer is voorzien van een polarografische zuurstofsensor. Cellulair of geïsoleerde mitochondriële schorsingen worden continu in de respirometer geroerd. Mitochondriale functie, substraten en remmers beoordelen op mitochondriale complex activiteit worden toegevoegd volgens standaard protocollen. Substraten en remmers kunnen worden getitreerd door injectie into de kamers van de oxygraph en zuurstofverbruik tarieven worden berekend met behulp van software en uitgedrukt als picomol per seconde per aantal cellen. Hoge-resolutie respirometrie biedt verschillende voordelen vergeleken met traditionele en gebruikelijke polarografische zuurstofelektrode inrichtingen waaronder verhoogde gevoeligheid en kunnen werken met kleine hoeveelheden biologische monsters zoals intacte of gepermeabiliseerde cellen. Bovendien, elke inrichting bevat twee kamers en ademfrequentie tegelijk worden opgenomen voor vergelijkingen van zuurstofconcentraties. De hoge-resolutie oxygraph heeft ook het voordeel van verminderde lekkage van zuurstof uit het apparaat kamers in vergelijking met traditionele polarografische zuurstof stroomdraden. Een ander apparaat recent ontwikkelde cellulaire zuurstofverbruik te meten, is de 96-well extracellulaire flux analyzer 5. Het extracellulaire flux analysator voorzien van fluorescentie in plaats van polarografische sensoren. De voordelen van de extracellulaireflux analyzer vergelijking met de hoge-resolutie oxygraph zijn i) het is een grotendeels geautomatiseerd middel, ii) kan zuurstofconsumptie meten 24- en 96-well platen voor high-throughput screening, daarvoor zijn kleinere hoeveelheden biologische monsters, en iii) aanvullende meting van cellulaire glycolytische flux mogelijk. De nadelen van de extracellulaire flux analysator in vergelijking met de hoge-resolutie oxygraph zijn i) de hoge kosten van het apparaat en verbruiksartikelen zoals fluorescerende platen, die niet herbruikbaar zijn en ii) slechts vier verbindingen per assay / putje injecteerbare daarom is het systeem niet haalbaar substraat-ontkoppelaar-inhibitor titratie protocollen. In de huidige studie, maken we gebruik van een hoge resolutie respirometrie mitochondriale ademhaling te bepalen. Voor cellulaire zuurstofverbruik experimenten worden de cellen permeabel gemaakt om de invoer van exogene ADP en mitochondriale oxideerbare substraten mogelijk maken toevoeren elektronen in complexes van de luchtwegen. Deze aanpak maakt het ontleden van afzonderlijke complexen mitochondriale respiratoire capaciteit, hetgeen een duidelijk voordeel in vergelijking met intacte cellen (vele substraten cel-impermeant). Echter celmembraan permeabilisatie de barrière tussen het cytosol en extracellulaire ruimte en medium (wassen cytosolisch opgeloste stoffen) en de samenstelling van de intracellulaire ruimte is geëquilibreerd met de extracellulaire ruimte verstoren. Een van de nadelen van gepermeabiliseerde cellen over intacte cellen die de mitochondriale buitenste membraan kan beschadigd raken als grote hoeveelheden reinigingsmiddel worden gebruikt tijdens cel permeabilisatie. In intacte cellen, basale, gekoppeld en ontkoppeld ademhaling van intacte cellen kan worden gemeten. Deze methode evalueert zuurstofverbruik van intacte cellen zonder toevoeging van exogene substraten en ADP, reproduceren de ademhalingsfunctie in de geïntegreerde cel en het leveren van gegevens over maximale mitochondriale electron transport capaciteit 6, 7. Een van de voordelen van intacte cellen in gepermeabiliseerde cellen die cellulaire omgeving niet verstoord en mitochondria in contact met de gehele componenten van de cellen. Om de cellulaire plasmamembraan permeabel zijn detergentia, zoals digitonine 8 gebruikt. Echter, mitochondriale buitenste membraan integriteit in het gedrang komen als grote hoeveelheden digitonine worden toegepast. Dat mitochondriale buitenste membraan integriteit niet in gepermeabiliseerde cellen aangetast bevestigen, wordt digitonine titratie uitgevoerd om de optimale concentratie voor mobiele permeabilisatie bepalen. Voor deze experimenten worden cellen opnieuw gesuspendeerd in medium ademhaling en digitonine concentratie wordt getitreerd met respirometrie in aanwezigheid van mitochondriale substraten en ADP en ademhalingssnelheden gemeten. Ademhaling van intacte, niet- gepermeabiliseerde cellen niet gestimuleerd in aanwezigheid van mitochondrial substraten en ADP. Toch zou daaropvolgende stapsgewijze digitonine titratie geleidelijke permeabilisatie van plasmamembranen opbrengst en optimale digitonine concentratie verkregen. Dit blijkt uit de toename van de ademhaling tot volledige permeabilisatie. Mitochondriale kwaliteit en buitenste membraan kunnen worden geverifieerd door het toevoegen van exogeen cytochroom c 2, 9. Cytochroom c is een 12 kDa eiwit elektronen uitvoering van de mitochondriale elektronentransportketen 10, 11, 12. Het is gelokaliseerd in het mitochondriaal intermembrane ruimte, en is betrokken bij zuurstofverbruik, dragende elektronen van complex III complex IV. Zodra de mitochondriale buitenste membraan beschadigd wordt cytochroom c vrijgegeven en mitochondriale zuurstofverbruik wordt verlaagd. Na toevoeging van exogene cytochroom c, geen verhoging in mitochondriale respiratie is indicatief voor eenverstoorde mitochondriale buitenmembraan. In gepermeabiliseerde cellen, substraten en remmers van mitochondriale complexe activiteit worden toegevoegd na diverse protocollen 3, 9. Bijvoorbeeld om mitochondriale complex aangedreven ademhalingssnelheid onderzoeken, kan het volgende protocol worden gebruikt. Na permeabilisatie van de cellen, wordt eerst complex I gestimuleerd door de substraten malaat en glutamaat, die NADH genereren als substraat om de ademhalingsketen en de activering van complexe I. Daarna ADP toegevoegd voor omzetting in ATP (state 3, actieve provoceren complex I-afhankelijke ademhaling). Nadat een stabiel signaal bereikt wordt rotenon (mitochondriale complex I-remmer) toegediend complexe I. Rotenone volgt succinaat tot FADH 2 remmen en complexe II (state 3, actieve complex II-afhankelijke ademhaling) activeren. Om complexe IV-afhankelijke ademhaling, eerste complex III metenafhankelijke ademhaling wordt geremd door toevoeging antimycin A (mitochondriale complex III inhibitor). Daarna wordt complex IV-afhankelijke ademhaling gestimuleerd door toediening van ascorbaat en tetramethylphenylendiamine (TMPD). TMPD kunnen auto-oxideren in de ademhaling buffer derhalve de maximale complex IV-afhankelijke ademhalingssnelheid (State 3) wordt berekend door ademhaling voor en na de toevoeging van natriumazide, een remmer van mitochondriale complex IV. De ademhaling experimenten uitgevoerd in twee kamers van een oxygraph parallel-één dient als een controle (niet-gestimuleerde cellen), de andere met de gestimuleerde cellen worden uitgevoerd. Uiteraard kunnen de cellen worden voorbehandeld verschillende wijzen, bijvoorbeeld met geneesmiddelen beïnvloeden mitochondriale functies voor hun zuurstofverbruik wordt gemeten in de kamer oxygraph. Dit protocol maakt functionele onderzoek van de individuele mitochondriale ademhalingsketen complexen. Bovendien kan men meten maximale ADP-gestimuleerdeademhaling (toestand 3) van gepermeabiliseerde cellen, middels exogeen vetzuur in de vorm van palmitaat. In dit protocol wordt geconcentreerd voorraden natriumpalmitaat geconjugeerd met ultra vetzuurvrij runderserumalbumine (BSA) (6: 1 molverhouding palmitaat: BSA). Daarna cellen eerst worden gepermeabiliseerd met digitonine en mitochondriale ademhaling wordt bepaald door het toevoegen van carnitine en palmitinezure gevolgd door toevoeging van ADP (state 3, maximale ademhaling). Vervolgens wordt oligomycin toegevoegd aan toestand 4 (state 4o) en respiratoire controle ratio (RCR-waarde) na te bootsen wordt berekend als toestand 3 / staat 4o. β-oxidatie bevordert de productie van acetyl CoA (die gaat in de TCA cyclus) en het genereren van FADH 2 en NADH, waarvan de elektronen aan de elektronentransport keten voorbij elektronen overdracht flavoproteïne en β-hydroxyacyl-CoA dehydrogenase. Mitochondriën zijn in het centrum van vetzuurmetabolisme en beschreven palmitaat-BSA protocol kan worden gebruikt door onderzoekers onderzoeken fatty zuur oxidatie. In intacte cellen, activatoren en remmers van mitochondriale complexe activiteit worden toegevoegd naar aanleiding van een ander protocol 6, 9. Voor deze experimenten wordt eerst zuurstofverbruik van niet-gepermeabiliseerde cellen in de afwezigheid van exogene substraten gemeten (fosforyleert ademhaling). Vervolgens wordt de niet-fosforylerende ademhalingssnelheid gemeten na toevoeging van oligomycin, dat een remmer van mitochondriale ATP synthase. Daarna wordt de protonophore carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) toegediend in verschillende concentraties en de maximale mitochondriale ontkoppelde ademhalingsfrequentie wordt gemeten. Protonophores zoals FCCP kan een verhoging in proton permeabiliteit van het binnenste membraan te induceren, waardoor passieve beweging van protonen aan de chemiosmotic verloop verdwijnen. Een toename van permeabiliteit proton ontkoppeld oxidatieve respiratie (geen ATP productie) en veroorzaakt een toename oxygen consumptie. Daarna rotenon en antimycin A toegevoegd aan mitochondriale ademhaling remmen en non-mitochondriale ademhaling wordt afgetrokken van alle andere ademhalingssnelheid. Het zuurstofverbruik kan worden uitgedrukt als IO 2 [pmol x sec -1 x 10 -6 cellen] (zuurstofstroom per miljoen cellen) die wordt berekend door het volume-specifieke zuurstofstroom (in de gesloten zuurstofkamer), JV, o 2 [pmol x sec -1 x ml -1] van cel-concentratie in de cel kamer (aantal cellen per volume [10 6 cellen ∙ ml 1]) 15. high-resolution respirometry to assess mitochondrial function in permeabilized and intact cells

A high-resolution oxygraph is a device for measuring cellular oxygen consumption in a closed-chamber system with very high resolution and sensitivity in biological samples (intact and permeabilized cells, tissues or isolated mitochondria). The high-resolution oxygraph device is equipped with two chambers and uses polarographic oxygen sensors to measure oxygen concentration and calculate oxygen consumption within each chamber. Oxygen consumption rates are calculated using software and expressed as picomoles per second per number of cells. Each high-resolution oxygraph chamber contains a stopper with injection ports, which makes it ideal for substrate-uncoupler-inhibitor titrations or detergent titration protocols for determining effective and optimum concentrations for plasma membrane permeabilization. The technique can be applied to measure respiration in a wide range of cell types and also provides information on mitochondrial quality and integrity, and maximal mitochondrial respiratory electron transport system capacity.
