Brûler les infections continuent d&#39;être une cause importante de morbidité et de mortalité. L&#39;émergence croissante des bactéries multirésistantes (MDR) a conduit à l&#39;échec fréquent des traitements antibiotiques traditionnels. D&#39;autres sont thérapeutiques d&#39;urgence nécessaires pour lutter contre les bactéries MDR. Une approche innovante non-antibiotique, bleu clair antimicrobien (ABL), a montré l&#39;efficacité prometteuse contre les infections MDR. Le mécanisme d&#39;action de ABL est pas encore bien compris. Il est généralement supposé que sont excités par ABL, qui à son tour se produisant naturellement chromophores photosensibilisants endogènes dans des bactéries (par exemple, les porphyrines libres de fer, les flavines, etc.) produit des espèces oxygénées réactives cytotoxiques (ROS) par l&#39; intermédiaire d&#39; un processus photochimique. Contrairement à une autre approche antimicrobienne à base de lumière, la thérapie photodynamique antimicrobien (aPDT), la thérapie ABL ne nécessite pas l&#39;intervention d&#39;un photosensitiz exogèneer. Tout ce qu&#39;il a besoin de prendre effet est l&#39;irradiation de la lumière bleue; , Il est donc simple et peu coûteuse. Les récepteurs ABL sont les photosensibilisants endogènes cellulaires chez les bactéries, plutôt que de l&#39;ADN. Ainsi, Abl est considérée comme beaucoup moins génotoxique à des cellules hôtes que ultraviolets C (UVC) irradiation, ce qui provoque directement des dommages de l&#39;ADN dans des cellules hôtes. Dans cet article, nous présentons un protocole pour évaluer l&#39;efficacité du traitement ABL pour MDR Acinetobacter baumannii infections dans un modèle de souris de brûlure. En utilisant une souche bioluminescente d&#39;ingénierie, nous avons pu surveiller de manière non invasive l&#39;étendue de l&#39;infection en temps réel chez les animaux vivants. Cette technique est également un outil efficace pour la surveillance de la distribution spatiale des infections chez les animaux.

La souche de A. baumannii que nous avons utilisé est un isolat clinique MDR, tel que rapporté précédemment 12, 17.
