Laser ablatie - inductief gekoppeld plasma - massaspectrometrie (LA-ICP-MS) beeldvorming maakt gebruik van een gefocusseerde UV-laserbundel die variëren in grootte van 1 tot&gt; 100 urn in diameter (of breedte bij een vierhoek bundelvorm wordt gebruikt), waarbij de monster afgelopen 7. Metalen worden een rode draad door een organisme gevonden, met hun biologische rol gedicteerd door zowel hun chemische reactiviteit en overvloed binnen een bepaalde anatomische regio. In de hersenen, metalen hebben een sterk gecompartimenteerd distributie, afhankelijk van de primaire functie zij binnen het centrale zenuwstelsel. Afbeelden van de ruimtelijke verdeling van metalen heeft unieke inzicht gegeven in de biochemische architectuur van de hersenen, waardoor directe correlatie tussen neuroanatomische regio en de bekende functie met betrekking tot metaal-afhankelijke processen. Daarnaast beschikken over verschillende leeftijd gerelateerde neurologische aandoeningen verstoorde homeostase metaal, vaak beperkt tot kleine gebieden van de hersenen die anders moeilijk te analyseren zijn. We beschrijven hier een uitgebreide werkwijze voor kwantitatief afbeelden metalen in muizenhersenen, middels laser ablatie - inductief gekoppeld plasma - massaspectrometrie (LA-ICP-MS) en speciaal ontworpen beeldverwerkingsoftware. Gericht op ijzer, koper en zink, waarbij drie van de meest voorkomende en ziekte-relevante metalen in de hersenen, de ter zake stappen monsterbereiding, analyse, kwantitatieve metingen en beeldverwerking kaarten metaal verdeling in de lage micrometer produceren beschrijven resolutie afstand. Deze techniek van toepassing op alle gesneden weefselsectie, kan tonen dat zeer variabele verdeling van metalen binnen een orgaan of systeem, en kunnen worden gebruikt om veranderingen in metaal homeostase en absolute niveaus binnen fijne anatomische structuren te identificeren.

Om de mogelijkheden van deze LA-ICP-MS imaging benadering tonen, een eenvoudig experiment met een enkel stuk WT C57BL / 6 muizenhersenen, doorsneden in het corpus callosum en deelbaar in het frontale vlak wordt gepresenteerd. Een werkstroom voor de analyse van gegevens met behulp Biolite (figuur 1), zoals beschreven in de paragrafen 6 en 7, en voor het verschaffen van een representatief beeld van metaaldistributie in de sectie analyse (figuur 2) is ook beschreven. Zoals blijkt, metaalverdeling in muizenhersenen varieert volgens anatomisch gebied. Dit kan worden toegeschreven aan de variabele rollen metalen, en meer bepaald de eiwitten waaraan ze gebonden zijn, spelen in elk hersengebied 27. Bijvoorbeeld ijzer meestal hoger concentraties in de middenhersenen en langs de dentate gyrus, terwijl zink is het meest overvloedig in de cortical gebieden. Koolstof, hetgeen een algemeen gebruikte interne standaard 8, homogeen verdeeld. Elemental kaarten (figuur 2) kan bijzonder nuttig zijn wanneer gebruikt in combinatie met bestaande anatomische en functionele referentie atlassen 29, waarbij gegevens over de colokalisatie van metalen kan de expressie van specifieke metaalbindende eiwitten kan inzicht in de functie van metalen in hersenen regio, of veranderingen in de metaal niveaus in lijn met een geïdentificeerde ziekte-gerelateerde biomolecuul. Met de beschreven benadering, die is geoptimaliseerd voor een breder scala analyten, uitsluit gevoeligheid voor lage abundantie elementen zoals mangaan en werkwijzen kunnen worden aangepast om voornamelijk richten op het analyt door het verhogen verblijftijden ten koste van andere gemeten massa. Het grote voordeel bij het gebruik van beeldvorming door LA-ICP-MS is het observeren van relatieve verschillen in metalen concentratie en de verdeling tussen de experimentele groepen. We hebben eerder gebruikte techniek om dergelijke verhoogde strijken na een insult neurotoxine nabootsen PD 2, 10, en veranderingen in corticale ijzer niveaus in Alzheimer weefsel 21 te tonen.
