Dit protocol beschrijft de vorming van blaastumoren bij vrouwelijke C57BL / 6J muizen, waarbij de muis blaaskanker cellijn MB49, dat gemodificeerd menselijk Prostaat Specifiek Antigeen (PSA) scheiden, en de procedure voor de bevestiging van tumor implantatie. In het kort worden muizen verdoofd met behulp van injecteerbare geneesmiddelen en zijn gemaakt te leggen in de dorsale positie. Urine wordt vrijgemaakt uit de blaas en 50 pl poly-L-lysine (PLL) wordt langzaam gedruppeld met een debiet van 10 ul / 20 s onder toepassing van een 24 G IV katheter. Het wordt in de blaas gedurende 20 min door stoppering de katheter. De katheter wordt verwijderd en PLL hebben verlaten lichte druk op de blaas. Dit wordt gevolgd door instillatie van het muizen blaaskanker cellijn (1 x 10 5 cellen / 50 ul) met een snelheid van 10 pl / 20 s. De katheter wordt afgesloten om voortijdige afvoer voorkomen. Na 1 uur worden de muizen herleven een ommekeer geneesmiddel en de blaas wordt ontruimd. De langzame indruppeling snelheid is belangrijk,aangezien er minder vesico-ureterale reflux, wat kan leiden tot tumoren kunnen optreden in het bovenste urinewegen en de nieren. De cellijn moet goed worden geresuspendeerd tot samenklontering van cellen te verminderen, aangezien dit kan leiden tot ongelijke tumorgrootten na implantatie. Deze techniek veroorzaakt tumoren met hoog rendement. De tumorgroei wordt gevolgd door PSA urinaire uitscheiding. PSA marker monitoring betrouwbaarder dan echografie of fluorescentie ter detectie van de aanwezigheid van tumoren in de blaas. Tumoren bij muizen in het algemeen te bereiken van een maximale grootte die een negatieve invloed op de gezondheid van ongeveer 3-4 weken indien onbehandeld. Door het monitoren tumorgroei, is het mogelijk om muizen die waren genezen van degenen die niet succesvol werden geïmplanteerd met tumoren onderscheiden. Met slechts eindpunt analyse, kan deze ten onrechte worden geacht te zijn genezen door therapie.

PSA secretie van MB49 cellen bleek te variëren met de groeimedia. MB49-PSA wordt gekweekt in DMEM media, omdat dit resulteert in verhoogde PSA uitscheiding (Figuur 1A).
