Dieses Protokoll beschreibt die Erzeugung von Blasentumoren in weiblichen Mäusen C57BL / 6J der murinen Blasenkrebs-Zelllinie MB49 verwendet, die menschliche Prostataspezifischen Antigen (PSA), und das Verfahren für den Nachweis der Tumorimplantation abzusondern modifiziert wurde. Kurz gesagt, werden die Mäuse mit injizierbaren Medikamenten betäubt und in der Rückenlage zu legen hat. Urin aus der Blase geräumt und 50 ul Poly-L-Lysin (PLL) wird langsam mit einer Geschwindigkeit von 10 &amp; mgr; l / 20 s unter Verwendung einer 24 G IV-Katheter instilliert. Es wird 20 min in der Blase links durch den Katheter Stopfen verschlossen. Der Katheter wird entfernt und PLL wird durch sanften Druck auf die Blase verlassen. Dies wird durch Instillation des murinen Blasenkrebs - Zelllinie (1 x 10 5 Zellen / 50 &amp; mgr; l) mit einer Geschwindigkeit von 10 &amp; mgr; l / 20 s folgte. Der Katheter wird zugestöpselt vorzeitige Entleerung zu verhindern. Nach 1 h werden die Mäuse mit einer Umkehrung Droge wieder auf, und die Blase verlassen wird. Die langsame Einträufeln Rate ist wichtig,wie es reduziert vesico-ureteral Reflux, die Tumoren in der oberen Harnwege und in den Nieren auftreten verursachen können. Die Zelllinie sollte auch wieder ausgesetzt werden Verklumpung von Zellen zu reduzieren, da dies zu einer ungleichmäßigen Tumorgrößen nach der Implantation führen kann. Diese Technik induziert Tumoren mit hohem Wirkungsgrad. Das Tumorwachstum wird durch Urin-PSA-Sekretion überwacht. PSA Marker Überwachung ist zuverlässiger als Ultraschall oder Fluoreszenzabbildung für den Nachweis der Anwesenheit von Tumoren in der Blase. Tumoren in Mäusen im Allgemeinen eine maximale Größe, die einen negativen Einfluss auf die Gesundheit von etwa 3 erreichen - 4 Wochen, wenn sie unbehandelt. Durch die Überwachung des Tumorwachstums ist es möglich, Mäuse zu unterscheiden, die von den ausgehärteten wurden, die nicht erfolgreich mit Tumoren implantiert. Mit nur Endpunktanalyse, kann der letztere fälschlicherweise angenommen, dass durch Therapie geheilt wurden.

PSA Sekretion von MB49-Zellen wurde festgestellt, mit dem Wachstumsmedium zu variieren. MB49-PSA wird in DMEM - Medium gezüchtet , weil dies zu einer erhöhten PSA - Sekretion (1A).
