Målet med denne metoden er å generere murine ortotopiske blæren svulster og for å overvåke de implanterte svulster så nøyaktig som mulig, slik at mus uten svulst implantasjon ikke er antatt å ha blitt kurert ved utgangen av punkt-analyse. Totalt sett, idet fremgangsmåten er vist vil redusere behovet for et stort antall mus for eksperimentell analyse og sikre større nøyaktighet ved bestemmelse terapeutiske resultater. Utviklingen av en ortotopisk modell for kreft er viktig, da implantere tumorceller subkutant ikke rekapitulere miljøet av klinisk sykdom eller muliggjøre utvikling av terapeutiske strategier. Arkitekturen av blæren tillater instillasjon av blære kreft terapi direkte inn i blæren med minimale systemiske effekter. Således dyremodeller som rekapitulere dette miljøet, slik som en ortotopisk modell, er viktig for evaluering av nye behandlingsformer. Den metode som er utviklet av Ninalga et al. benytter den positivt ladede poly-L-lysin (PLL) 6 molekyler å belegge blæreveggen; Dette gjør at de negativt ladede kreftceller å holde seg til glycosaminoglycan lag av blæren. Denne fremgangsmåte resulterer generelt i mer enn en tumor utvikling i blæren, men tumorimplantering er ved 80 - 100% 4,7. Teknisk sett er det også den enkleste fremgangsmåten for å utføre. For å sikre at de tumorer som utvikler seg er ganske selv i størrelse, er det viktig at tumorcellene ikke er gruppert i store klumper før implantasjon. Dermed vil en god deteksjonssystem som kan måle tumorstørrelse kort tid etter implantering er viktig. Flere strategier har been brukes til å evaluere tumorer. Disse omfatter magnetisk resonansavbildning (MRI) 8-10, fluorescens 11, bioluminescens 12,13, ultralyd 14, og enzym-bundet immunosorbent assay (ELISA) 15,16. Mens MRI og ultralyd ikke krever modifiseringer av tumorceller, er det behov for følsomme utstyr og kontrastmidler for MRI. De fluorescence-, luminescence-, og ELISA-baserte analyser krever endring av kreftceller til å uttrykke markørproteiner som kan oppdages av disse metodene. For luminescens, er et substrat som er nødvendig for påvisning av luciferase-aktivitet; således er det et ekstra trinn og økte kostnader. Både luminescens og fluorescens krever spesialisert utstyr. Denne protokollen beskriver dannelse av blæretumorer i hunn C57BL / 6J-mus ved bruk av murine blærecancercellelinje MB49, som har blitt modifisert til å utskille human prostata spesifikt antigen (PSA), og fremgangsmåten for bekreftelse av tumorimplantering. Kort fortalt blir musene bedøvet bruker injiserbare legemidler og er laget for å ligge i ryggposisjon. Urin forlates fra blæren og 50 ul av poly-L-lysin (PLL) blir langsomt tildryppet med en hastighet på 10 ul / 20 s ved anvendelse av en 24 G IV-kateter. Det er igjen i blæren i 20 min ved å legge stopper kateteret. Kateteret fjernes og PLL er forlatt med lett press på blæren. Dette etterfølges av instillasjon av den murine blærecancercellelinje (1 x 10 5 celler / 50 ul) med en hastighet på 10 pl / 20 s. Kateteret er tettet for å hindre for tidlig evakuering. Etter 1 time blir musene gjenopplivet med en reversering narkotika, og blæren er forlatt. Den langsomme instillasjon hastighet er viktig,da det reduserer vesico-urinleder tilbakeløp, noe som kan føre til svulster å forekomme i den øvre urinveiene og i nyrene. Den cellelinje bør være godt resuspenderes å redusere klumping av celler, da dette kan føre til ujevnheter i tumorstørrelser etter implantasjon. Denne teknikken induserer tumorer med høy effektivitet. Tumorvekst blir overvåket ved urin PSA sekresjon. PSA markør overvåkning er mer pålitelig enn ultralyd eller fluorescens avbildning for påvisning av nærvær av tumorer i blæren. Svulster i mus generelt nå en maksimal størrelse som negativt påvirker helsen ved ca 3-4 uker hvis venstre ubehandlet. Ved å overvåke tumorvekst, er det mulig å differensiere mus som ble herdet fra de som ikke var fått implantert med tumor. Med bare ende-punkt-analyse, kan den sistnevnte være feilaktig antas å ha blitt herdet ved terapi.

PSA-sekresjon fra MB49-celler ble funnet å variere med vekstmediet. MB49-PSA blir dyrket i DMEM-media, fordi dette resulterer i økt PSA sekresjon (figur 1A).
