Sinds de introductie van Medawar, eerst in menselijke studies en bij konijnen en muizen is huidtransplantatie een waardevol model voor de studie van allogene immune responsen 6,7 geweest. In dit manuscript presenteren we een model van grootschalige, niet-gevasculariseerd, full-thickness huidtransplantatie met de bovenste en onderste rugvel. Orgaantransplantatie is de behandeling van keuze voor patiënten met eindstadium orgaanfalen en resultaten zijn opmerkelijk verbeterd met vooruitgang in chirurgische procedures en immunosuppressie protocollen. Echter langdurige immunosuppressie gepaard met significante bijwerkingen, en de ontwikkeling van nieuwe strategieën verdraagzaamheid blijft het doel van de moderne transplantatieonderzoek. Talrijke diermodellen zijn ontwikkeld voor basisonderzoek bij transplantatie, de mechanismen van transplantaatafstoting bestuderen en te testen immunosuppressie benaderingen voor het voorkomen van transplantaat afstoting en voor de bevordering van tolerantie op lange termijn 1-3. Muismodellen hebben de steunpilaar van immunologisch onderzoek geworden als gevolg van de exclusieve en ruime beschikbaarheid van diagnostische en therapeutische antilichamen en goed gedefinieerde inteelt en transgene stammen. Huidtransplantatie is een eenvoudige procedure die niet vereist speciale vaardigheden en kan microchirurgischegemakkelijk postoperatief gevolgd. Bij elkaar genomen, heeft de muis huidtransplantatie een uitzonderlijk hulpmiddel om vele aspecten die betrokken zijn bij de alloimmuun reactie, met inbegrip van antigeen-afgifte, mobiele handel, en weefsel tijdens afstoting van 4,5 te bestuderen. Hier tonen we de stap-voor-stap procedure voor full-thickness huidtransplantatie met het muismodel en we de belangrijke stappen die nodig zijn voor een succesvolle implantatie van de getransplanteerde huid beschrijven.

murine full-thickness skin transplantation

Murine volledige dikte huidtransplantatie is een gevestigde in vivo model om allo respons en transplantaatafstoting bestuderen. Ondanks de beperkte toepassing voor mensen, is huidtransplantatie bij muizen grote schaal toegepast voor transplantatie onderzoek. De procedure is eenvoudig te leren en uit te voeren, en het vereist geen delicate microchirurgische technieken, noch uitgebreide training. Bovendien transplantaatafstoting in dit model wordt op een zeer reproduceerbare immunologische reactie en wordt gemakkelijk gevolgd door directe inspectie en palpatie. Bovendien kunnen secundaire huidtransplantatie met donor gematched of skin derden grafts uitgevoerd complexere transplantatie modellen als alternatief en eenvoudige methode om donor-specifieke tolerantie te bepalen. De complicaties zijn laag en zijn over het algemeen beperkt tot anesthesie overdosering of ademhalingsproblemen na de ingreep. Transplantaatfalen, daarentegen, komt vaak voor als gevolg van slechte voorbereiding van de grachterschip, onjuiste positionering in het transplantaat bed, of onjuiste plaatsing van de bandage. In dit artikel presenteren we een protocol voor volledige dikte huidtransplantatie bij muizen en beschrijf de belangrijke stappen die nodig zijn voor een succesvolle procedure.
