Sin embargo, la inmunosupresión a largo plazo se asocia con efectos secundarios significativos, y el desarrollo de nuevas estrategias que promueven la tolerancia sigue siendo el objetivo de la investigación trasplante moderna. Numerosos modelos animales se han desarrollado para la investigación básica en el trasplante, para el estudio de los mecanismos de rechazo de aloinjertos y para poner a prueba la inmunosupresión se aproxima a la prevención del rechazo del injerto y para promover la tolerancia a largo plazo 1-3. Los modelos de ratón han convertido en el pilar de la investigación inmunológica debido a la disponibilidad exclusiva y extensa de anticuerpos terapéuticos y de diagnóstico y cepas puras y transgénicos bien definidos. El trasplante de piel es un procedimiento simple que no requiere conocimientos especiales de microcirugía y puedemonitorizar fácilmente después de la operación. En conjunto, el trasplante de piel de ratón ha sido una herramienta excepcional para estudiar muchos aspectos implicados en la respuesta aloinmune, incluyendo la entrega de antígeno, el tráfico de células y la destrucción del tejido durante el rechazo de injerto 4,5. murine full-thickness skin transplantation

Murino de trasplante de todo el espesor de la piel es un modelo in vivo bien establecido para estudiar la respuesta aloinmune y rechazo del trasplante. A pesar de su aplicación limitada a los seres humanos, el trasplante de piel en ratones ha sido ampliamente empleado para la investigación del trasplante. El procedimiento es fácil de aprender y realizar, y que no requiere técnicas de microcirugía delicada ni una amplia formación. Además, el rechazo del injerto en este modelo se produce en una reacción inmunológica muy reproducible y se monitoriza fácilmente mediante la inspección directa y palpación. Además, el trasplante de piel secundaria con injertos de donantes emparejados o de la piel de otro fabricante puede llevar a cabo en modelos de trasplante más complejos como un método alternativo y sin complicaciones para evaluar la tolerancia específica del donante. Las complicaciones son bajos y son, en general, limitada a una sobredosis de anestesia o dificultad respiratoria después del procedimiento. El fracaso del injerto, por el contrario, se produce habitualmente como resultado de una mala preparación de la grpopa, posicionamiento incorrecto en el lecho del injerto, o la colocación inadecuada de la venda. En este artículo, se presenta un protocolo para el trasplante de piel de espesor completo en ratones y describimos los pasos son importantes y necesarias para el éxito del procedimiento.

La colocación del vendaje en el ratón receptor es un paso importante del procedimiento. El injerto de piel se coloca en el tronco receptor, entre el hombro, cadera y columna vertebral (Figura 1).
