Sedan introduktionen av Medawar, först i humanstudier och sedan i kaniner och möss har hudtransplantation varit en ovärderlig modell för studier av allogena immunsvar 6,7. Organtransplantation är behandling av valet för patienter med gravt organsvikt, och resultaten har förbättrats avsevärt med framsteg inom kirurgiska ingrepp och immunosuppression protokoll. Dock är långsiktigt immunosuppression i samband med betydande biverkningar, och utveckling av nya strategier som främjar tolerans förblir målet för moderna transplantationsforskning. Ett stort antal djurmodeller har utvecklats för grundforskning vid transplantation, för att studera mekanismerna bakom allograftavstötning och testa immunsuppression metoder för att förhindra avstötning av transplantat och för att främja långtidstolerans 1-3. Musmodeller har blivit grunden för immunologisk forskning på grund av den exklusiva och stora tillgången på diagnostiska och terapeutiska antikroppar och väldefinierade inavlade och transgena stammar. Hudtransplantation är en enkel procedur som inte kräver speciella mikrokunskaper och kanlätt övervakas postoperativt. Sammantaget har mushud transplantation varit ett utomordentligt verktyg för att studera många aspekter på alloimmun svar, bland annat antigen leverans, cell trafficking, och vävnadsdöd under avstötning 4,5. Här visar vi steg-för-steg förfarande för full tjocklek hudtransplantation använder musmodell och vi beskriver de viktiga åtgärder som krävs för en lyckad engraftment av transplanterade hud.

murine full-thickness skin transplantation

Murin full tjocklek hudtransplantation är en väletablerad in vivo modell för att studera alloimmun svar och avstötning av transplantat. Trots sin begränsade tillämpning på människor, har hudtransplantation på möss i stor utsträckning använts för transplantation forskning. Förfarandet är lätt att lära och utföra och det kräver inte känsliga mikrokirurgisk teknik eller omfattande utbildning. Dessutom, transplantatavstötning i denna modell förekommer i en mycket reproducerbar immunologisk reaktion och övervakas lätt genom direkt inspektion och palpation. Dessutom kan sekundär hudtransplantation med givare matchade eller tredjeparts hudtransplantation utföras på mer komplexa transplantationsmodeller som ett alternativ och okomplicerad metod för att bedöma givarspecifik tolerans. Komplikationerna är låga och är i allmänhet begränsade till anestesi överdos eller andnöd efter ingreppet.
