OI est un outil très utile, rapide et peu coûteux pour l&#39;imagerie moléculaire in vivo non invasive dans la recherche préclinique. Une force particulière d&#39;OI est la capacité de surveiller des processus hautement dynamiques comme les réponses inflammatoires. En outre, OI permet de suivre le cours d&#39;une maladie pendant une période prolongée, allant de jours en semaines. OI présente plusieurs avantages par rapport à d&#39;autres modalités d&#39;imagerie...

Les maladies auto-immunes telles que la polyarthrite rhumatoïde (RA) ou le psoriasis vulgaire sont classées comme des réactions d&#39;hypersensibilité de type retardé (DTHR). 1 La PR est une maladie auto-immune commune caractérisée par une synovite érosive et une destruction articulaire. 2 Les articulations arthritiques inflammées démontrent l&#39;infiltration et la prolifération de cellules inflammatoires, une expression accrue de cellules pro-inflammatoires conduisant à la formation de pannus, au cartilage et aux destructions osseuses. 3 , 4 Le clivage de molécules de matrice extracellulaire, comme le collagène par métalloproteinases matricielles (MMP), est essentiel pour la conversion tissulaire et l&#39;angiogenèse et provoque des destructions tissulaires. 5 , 6 Les réactions d&#39;hypersensibilité au contact (CHR) sont caractérisées par l&#39;agrégation des neutrophiles conduisant à un éclatement oxydatif. 7 Similaires à la RA, les MMP en CHR sont involontairesDans la conversion des tissus, la migration cellulaire et l&#39;angiogenèse afin d&#39;établir une inflammation chronique. Pour étudier la PR, on a utilisé le modèle de souris à injection injectable de glucose-6-phosphate isomérase (GPI) -serum. 8 Le sérum provenant de souris K / BxN transgéniques contenant des anticorps contre GPI a été injecté dans des souris BALB / c naïves après quoi une inflammation rhumatismale a commencé à se développer dans les 24 h avec un gonflement maximal de la cheville au jour 6 après une injection de GPI-sérum (voir 1.1). Pour analyser la CHR chronique, les souris C57BL / 6 ont été sensibilisées avec du trinitrochlorobenzène (TNCB) sur l&#39;abdomen. L&#39;oreille droite a été remise en question jusqu&#39;à 5 fois, commençant 1 semaine après la sensibilisation (voir aussi 1.1 et 1.2). L&#39;OI de petit animal non invasif est une technique basée sur l&#39;étude in vivo des signaux fluorescents, chimioluminescents et bioluminescents, qui sont principalement utilisés dans la recherche préclinique. Les données semi-quantitatives acquises donnent des indications sur la moléculeDans les organes et les tissus des modèles animaux expérimentaux sains et malades, et permet des mesures longitudinales de suivi ( par exemple pour évaluer les profils de réponses thérapeutiques in vivo ). Un grand avantage des études longitudinales est la réduction du nombre d&#39;animaux, car les mêmes animaux peuvent être mesurés dans des études de suivi à plusieurs moments au lieu d&#39;utiliser différentes souris par point de temps. La résolution de l&#39;OI permet une imagerie fonctionnelle détaillée des organes et même des structures tissulaires plus petites dans les animaux expérimentaux. L&#39;utilisation de filtres d&#39;excitation et d&#39;émission spécifiques avec un spectre de transmission étroit, une protection contre la lumière dispersée par une &quot;boîte foncée&quot; résistant à la lumière et une caméra de dispositif à couplage chargé (CCD) sensible qui est refroidie dans de nombreux appareils jusqu&#39;à -70 ° C , Permet des mesures très spécifiques et sensibles des signaux de fluorescence. En utilisant des agents fluorescents à excitation etLes spectres d&#39;émission dans la fenêtre de fluorescence proche infrarouge (650 - 950 nm), les rapports signal sur bruit peuvent être considérablement améliorés. La fenêtre de fluorescence proche du rayon infrarouge se caractérise par une absorption relativement faible du signal par l&#39;hémoglobine et l&#39;eau ainsi que par une faible fluorescence d&#39;arrière-plan. 9 Cela permet une profondeur de pénétration allant jusqu&#39;à 2 cm dans le tissu des petits animaux. Les sondes OI peuvent traiter directement une cible ( par exemple par un anticorps marqué par fluorescence) ou peuvent être activées dans le tissu cible ( par exemple par des protéases). Les sondes OI activables sont optiquement silencieuses sous leur forme inactivée en raison du transfert d&#39;énergie de résonance de Förster (FRET) à une fraction de trempe qui transfère l&#39;énergie d&#39;excitation dans la molécule à un autre domaine. Si le colorant est clivé (par une protéase par exemple), l&#39;énergie n&#39;est plus transférée dans la molécule et un signal fluorescent peut être détecté par OI. Cela permet de concevoir des sondes OI avec une spécificité élevéeY pour des processus biologiques distincts et d&#39;excellents rapports signal / bruit. Le protocole suivant explique en détail la préparation des animaux, les mesures OI en utilisant une sonde activable OI pour imaginer l&#39;activité MMP-2, -3, -9 et -13 in vivo et deux modèles expérimentaux d&#39;inflammation (RA, CHR).

non-invasive in vivo fluorescence optical imaging of inflammatory mmp activity using an activatable fluorescent imaging agent

Cet article décrit une méthode non invasive pour imager l&#39;activité de la métalloproteinases matricielles (MMP) par une sonde fluorescente activable, via l&#39; imagerie optique fluorescente in vivo (OI), dans deux modèles d&#39;inflammation de souris: une polyarthrite rhumatoïde (RA) et un contact Modèle de réaction d&#39;hypersensibilité (CHR). La lumière avec une longueur d&#39;onde dans la fenêtre infrarouge proche (NIR) (650 - 950 nm) permet une pénétration plus profonde des tissus et une absorption minimale du signal par rapport aux longueurs d&#39;onde inférieures à 650 nm. Les principaux avantages de l&#39;OI de fluorescence sont qu&#39;il est bon marché, rapide et facile à mettre en œuvre dans différents modèles animaux. Les sondes fluorescentes activables sont silencieuses optiquement dans leurs états inactivés, mais deviennent très fluorescentes lorsqu&#39;elles sont activées par une protéase. Les MMP activées conduisent à la destruction des tissus et jouent un rôle important pour la progression de la maladie dans des réactions d&#39;hypersensibilité à retard de type (DTHR) telles que la PR et la CHR. En outre, les MMP sontles protéases essentielles pour le cartilage et la dégradation des os et sont induites par les macrophages, les fibroblastes et les chondrocytes en réponse à des cytokines pro-inflammatoires. Ici, nous utilisons une sonde qui est activé par les MMPs clés comme la MMP-2, -3, -9 et -13 et décrivons un protocole d&#39;imagerie à fluorescence infrarouge proche OI de l&#39;activité de MMP dans la polyarthrite rhumatoïde et les souris témoins 6 jours après l&#39;induction de la maladie ainsi comme chez la souris avec aiguë (1x challenge) et chroniques (5x challenge) CHR sur l&#39;oreille droite par rapport à l&#39;oreille en bonne santé.

Pour induire la polyarthrite rhumatoïde (RA) chez des souris naïves BALB / c, les animaux ont été injectés ip avec des auto-anticorps (dilution 1: 1 avec 1x PBS) contre GPI au jour 0. L&#39;inflammation maximale (gonflement de la cheville) dans ce Sérum GPI induit Le modèle RA est le jour 6 après injection 11 .
