Un souci constant de NMDD est le ciblage contrôlé et la fourniture de formulations de médicaments à des cellules et des tissus, associée à une amélioration simultanée efficacité du médicament. Une classe spécifique de molécules de médicaments pour lesquels cela a posé un défi important est hydrophobes agents médicaments / d&#39;imagerie qui ont peu ou pas de solubilité dans les milieux aqueux. Depuis l&#39;avènement de l&#39;interfaçage nanomatériaux (matériaux ≤100 nm dans au moins une dimension) avec des cellules vivantes, un objectif constant a été de tirer parti des propriétés de taille dépendant uniques des nanoparticules (NPs) pour diverses applications. Ces applications comprennent l&#39; étiquetage cellulaire et tissulaire / imagerie (in vitro et in vivo), la détection en temps réel, et la livraison contrôlée de médicaments et d&#39; autres cargaisons 1. Des exemples de ces propriétés NP pertinents comprennent l&#39;émission dépendant de la taille des nanocristaux semi-conducteurs (quantum dots, QD); les propriétés photothermiques de nanoparticules d&#39;or; la grande capacité de chargement du coeur aqueux des liposomes; et la conductivité balistique des allotropes de carbone, tels que les nanotubes de carbone à paroi unique et graphène. Ceci est venu à être connu comme la délivrance de médicaments NP-médiée (NMDD), et il y a actuellement sept formulations de médicaments NP approuvés par la FDA pour une utilisation dans la clinique pour traiter divers cancers et des centaines d&#39;autres à divers stades d&#39;essais cliniques. La libération contrôlée d&#39;agents de médicament / d&#39;imagerie vers les cellules est critique pour le développement de thérapies et pour l&#39;étude des processus de signalisation cellulaire. Récemment, des nanoparticules (NP) ont montré prometteur dans le développement de ces systèmes de livraison. Ici, un NP (LCNP), système de délivrance à base de cristaux liquides a été utilisé pour la distribution contrôlée d&#39;un colorant insoluble dans l&#39;eau, le 3,3&#39;-perchlorate dioctadecyloxacarbocyanine (DIO), à l&#39;intérieur du noyau NP dans la région hydrophobe d&#39;un plasma bicouche membranaire. Lors de la synthèse des PN, le colorant est efficacement incorporé dans le coeur de LCNP hydrophobe, tel que confirmé par analyse spectroscopique multiples. Conjugaison d&#39;un dérivé du cholestérol pégylé à la surface NP (DiO LCNP-PEG-Chol) a permis la liaison du PN chargées de colorant sur la membrane plasmatique dans des cellules HEK 293T / 17 cellules. Résolue en temps laser microscopie confocale et Förster transfert d&#39;énergie par résonance (FRET) imagerie a confirmé le passageive efflux de DiO du noyau LCNP et son insertion dans la bicouche de la membrane plasmique. Enfin, la livraison de DiO comme LCNP-PEG-Chol atténué la cytotoxicité de DiO; la forme NP de DiO présentait ~ 30-40% moins de toxicité par rapport à DiO libre délivré de solution en vrac. Cette approche démontre l&#39;utilité de la plate-forme LCNP comme modalité efficace pour la prestation spécifique à membrane et la modulation des cargaisons moléculaires hydrophobes.
