מטרת המחקר הנוכחי הייתה לפתח טומוגרפיה טומוגרפיית פליטת פוזיטרונים / ממוחשבת (PET / CT) המתודולוגיה מבוססת לכמת את in vivo, בזמן אמת ספיגת הגלוקוז מזרם הדם לתוך רקמות ספציפיות בעכברים. זו הושגה באמצעות 18 F-שכותרתו fluorodeoxyglucose (FDG) כדי למדוד ספיגת הגלוקוז דוגמנות הקינטית כדי לאמוד את שיעורי 18 ספיגת F-FDG מן הפלזמה למרחב התאי, שיעור התחבורה מהחלל תאי לפלזמה והשיעור 18 זירחון F-FDG. במכרסמים, 18 F-FDG נעשה שימוש בהערכת טרום הקליני של טיפולים בסרטן רבים 1, מחקרים של התקדמות גידול 2 וחילוף חומרי גידול 3 וכן הדמיה של מאגרי שומן חום 4, neuroinflamation 5 והמוח המטבוליזם 6 </sup&gt;. שיטות מסורתיות להשתמש כדי לבחון את ספיגת רקמות ספציפיות של גלוקוז בעכברים (וחולדות) כרוך בדרך כלל לטיפול רדיואקטיבי 2-deoxyglucose גם עם 3 H או 14 C ואחריו המתת חסד, אוסף רקמות ומדידה של רדיואקטיביות בכל רקמות 7. שימוש PET / CT מאפשר קביעה פולשנית של ספיגת הגלוקוז וחילוף חומרים באיברים ואזורים בו זמנית בבעלי חיים. בנוסף, כמו המתת חסד אינה דרישה, טכניקה זו מתאימה לשימוש במחקרי אורך. סוכרת מסוג 2 (T2DM) מאופיינת חילוף החומרים של הגלוקוז מופרת היפרגליקמיה משנית היענות הרקמה מופחת אינסולין (תנגודת לאינסולין) וחוסר היכולת של -cells הלבלב לייצר כמות מספקת של אינסולין 8. ניתוח קינטי של ספיגת והגלוקוז יכול לספק תובנות חשובותמנגנון הפעולה ויעילות של התערבויות טיפוליות כמו גם לאפשר ניטור מתקדם של התקדמות מחלה.

determining glucose metabolism kinetics using 18f-fdg micro-pet/ct

This paper describes the use of 18F-FDG and micro-PET/CT imaging to determine in vivo glucose metabolism kinetics in mice (and is transferable to rats). Impaired uptake and metabolism of glucose in multiple organ systems due to insulin resistance is a hallmark of type 2 diabetes. The ability of this technique to extract an image-derived input function from the vena cava using an iterative deconvolution method eliminates the requirement of the collection of arterial blood samples. Fitting of tissue and vena cava time activity curves to a two-tissue, three compartment model permits the estimation of kinetic micro-parameters related to the 18F-FDG uptake from the plasma to the intracellular space, the rate of transport from intracellular space to plasma and the rate of 18F-FDG phosphorylation. This methodology allows for multiple measures of glucose uptake and metabolism kinetics in the context of longitudinal studies and also provides insights into the efficacy of therapeutic interventions.
