In de epidermis, immunohistochemie is een efficiënt middel te lokaliseren specifieke eiwitten van hun relatieve uitdrukking compartiment, namelijk de basale, suprabasale en stratum corneum lagen. De precieze lokalisatie binnen de epidermis van een bepaald eiwit geeft aanwijzingen in de richting van zijn functionele rol binnen de epidermis. In dit hoofdstuk beschrijven we een betrouwbare methode voor immunolocalization binnen de opperhuid aangepast voor zowel diepvries-en paraffine secties die we zeer regelmatig gebruiken in ons laboratorium. Paraffinecoupes over het algemeen een veel betere morfologie, dus betere resultaten en foto&#39;s, maar niet alle antilichamen zal samenwerken met de harde fixatie en de behandeling die betrokken zijn bij de verwerking ervan. Daarom is het protocol voor bevroren snijden ook. Binnen de paraffine snijden, zijn twee fixatie-protocollen beschreven (Bouin&#39;s en paraformaldehyde), de keuze van fixeermiddel zal direct worden gerelateerd aan het antilichaam specificaties en kan een andere bevestigingsmethode vereisen.
