I överhuden är immunohistokemi ett effektivt sätt att lokalisera specifika proteiner till deras relativa uttryck fack, nämligen den basala, suprabasal och hornlagret lager. Den exakta lokalisering inom epidermis av ett visst protein ger ledtrådar mot dess funktionella roll i epidermis. I detta kapitel beskriver vi en tillförlitlig metod för immunolocalization inom epidermis modifierats för både fryst och paraffin som vi använder mycket rutinmässigt i vårt laboratorium. Paraffin ger i allmänhet mycket bättre morfologi, alltså, bättre resultat och fotografier, men kommer inte alla antikroppar arbeta med de hårda fixering och behandling engagerade i sin behandling. Därför är protokollet för fryst sektionering också. Inom paraffin sektionering, finns två fixering protokoll som beskrivs (Bouins och paraformaldehyd), val av fixativ kommer att vara direkt relaterad till antikroppen specifikationer och kan kräva en annan fastställande metod.
