Het is een voorbeeld van een complex morfogenetisch proces waardoor meercellige gedrag optreden coördinerend de morfogenese op het niveau van afzonderlijke weefsels genereren. Deze differentiële weefsel vervormingen dan samenwerken om de verlenging van het achterste lichaam te genereren in de hele structuur niveau. Om te begrijpen hoe deze processen worden aangestuurd en gecoördineerd tijdens de ontwikkeling, moeten we in staat zijn om deze processen op verschillende schalen te volgen (dat wil zeggen op het niveau van moleculen, cellen, celpopulaties en weefsels) en deze rechtstreeks verband houden met de morfogenese van de gehele structuur . Zebravis embryo&#39;s zijn ideaal voor imaging achterste lichaam rek als hun optische transparantie en de geringe afmetingen zorgt voor de toepassing van minimaal invasieve licht imaging benaderingen zeer geschikt voor live beeldvorming. 1 Dit werd aangetoond door een aantal recente publicaties die gebleken werpen op achterste lichaam ontwikkeling op moleculair niveau, 2 enkele cellen, 3 en inter-weefsel gedrag, 4 en op het niveau van de celpopulatie en hele organen. 5 Geavanceerde imaging technieken zoals confocale, multi-foton microscopie en selectieve vliegtuig verlichting microscopie (SPIM) worden waardoor de lange termijn beeldvorming van ontwikkelingsprocessen met een verminderde werking van het licht toxiciteit en foto-bleken. Robuuste technieken voor de bevestiging van levende monsters nodig heeft drie doelen: 1) voldoende steun aan de bemonsteringen in de juiste positie te houden gedurende de overbrenging naar de microscoop en tijdens acquisitie, 2) voldoende bewegingsvrijheid van het monster om de uitgroei van het achterste lichaam regio zonder dat zijn normale ontwikwikkeling, en tenslotte 3) een zekere mate van veelzijdigheid van de montage-methode om imaging staan ​​op diverse imaging set-ups. Dit protocol introduceert een bevestigingstechniek voor het afbeelden van de ontwikkeling van de zebravis D. rerio. Deze techniek houdt montage embryo zodanig dat de kop en dooierzak regio vrijwel volledig opgenomen in agarose, terwijl het achterste lichaamsgebied te rekken en normaal te ontwikkelen. Zebravisembryo&#39;s vormen een ideale experimenteel systeem om complexe morfogenetische processen te bestuderen vanwege hun gemakkelijke toegankelijkheid en optische transparantie. Vooral achterste lichaam rek een essentieel proces in embryonale ontwikkeling waarbij meerdere weefsel vervormingen werken samen om de vorming van een groot deel van de lichaamsas sturen. Om dit proces te observeren door langdurige time-lapse imaging moet een bevestigingstechniek waarmee voldoende steun monsters in de juiste positie te houden gedurende de overbrenging naar de microscoop en verkrijging gebruiken. Daarnaast moet de montage ook voldoende bewegingsvrijheid voor de uitgroei van de achterste lichaamsgebied zonder aantasting van de normale ontwikkeling. Tenslotte moet een zekere veelzijdigheid van de montagewijze beeldvormende worden vrijgemaakt voor diverse imaging setups. Hier presenteren we een bevestigingstechniek voor beeldvorming van de ontwikkeling van achterste lichaam elongation in de zebravis D. rerio. Deze techniek houdt montage embryo zodanig dat de kop en dooierzak regio vrijwel volledig opgenomen in agarose, terwijl de achterste lichaamsgebied te rekken en normaal te ontwikkelen. We zullen zien hoe dit kan worden aangepast voor rechtop, omgekeerd en verticale light-sheet microscopie set-ups. Hoewel dit protocol is gericht op montage embryo&#39;s voor beeldverwerking met het achterste lichaam, kan het gemakkelijk worden aangepast voor de live imaging meerdere aspecten van zebravis ontwikkeling.
