There are three critical steps to successfully measure the LAV, Aorta and PA diameters. During the Set-up it is important to completely remove the fur on the chest; failure to do so will result in interference with image quality. During the Image Acquisition steps it is important to obtain each image with their corresponding probe and filter as the frames per second vary from probe to probe: that is 25 MHz and Cardiac Filter for the assessment of the LAV and 30 MHz and Abdominal Filter for assessment of the Aorta and PA ...

Den nåværende metoden lærer etterforsker hvordan å måle og bruke LAV, aorta diameter og PA diameter med 2D ekkokardiografi hos mus etter isofluran. Hjertesvikt med bevart ejeksjonsfraksjon (HFpEF) hos eldre mennesker rammer opp til 10% i de 80 år og eldre 1 og resulterer i betydelig sykelighet og dødelighet. Betydelig dødelighet oppstår også fra pulmonal hypertensjon (PH), en snikende sykdom prosess presenterer med lignende symptomer som HFpEF, hvor forhøyede lungearterien press føre til anstrengelsesdyspné, progressive høyre hjertesvikt, og ofte død. 2 Den økende utbredelsen av både HFpEF og PH betegne behovet for å utvikle en fremgangsmåte som gjør det mulig for nøyaktig evaluering og overvåking av intervensjoner i murine modeller i en ikke-invasiv, ikke-terminal tilnærming. Aldring fører til en forverring av diastolisk funksjon via endringer i ventrikkel-arterial stivne, vaskulær dysfunksjon, betennelser 3, nedsatt kalsium regulering 4, redusert β-adrenerge respons, og dårlig allmenntilstand produsere bremset aktiv avslapping og økt passiv stivhet. Den nåværende metoden lærer etterforsker hvordan å måle og bruke LAV som et surrogat for kronisk forhøyede venstre ventrikkels diastoliske trykket gjennom ekkokardiografi, samt å foreta målinger av aorta og PA diameter i mus. Mus som er eldre enn ti d av alder kan analyseres ved hjelp av den foreliggende teknikk. Teknikken består av 3 hovedtrinn: set-up, image oppkjøpet, og bildeanalyse. Oppsettet trinnet består av å få mus bedøvet med 1% isofluran, barbering det, og taping det i liggende stilling til et oppvarmet EKG bord hvor bildet oppkjøpet vil finne sted. Bildet oppkjøpet trinnet består av å lære å identifisere hjerte strukturer og skaffe alle de nødvendige bildene med sin korrespondent sonde og aksen for å kunne beregne volum og diameter.
