Het toenemende belang van magnetic resonance imaging (MRI) in klinische diagnose, die wordt begrensd door de inherente gevoeligheid van de techniek heeft geleid tot de snelle groei van het onderzoek naar de ontwikkeling van nieuwe gadolinium gebaseerde contrastmiddelen (GBCAs) 1. GBCAs zijn moleculen die worden toegediend om de beeldkwaliteit te verbeteren, en zij hebben meestal de chemische structuur van een trivalent ion gadolinium (Gd 3+) gecoördineerd aan een meertandige ligand. Dit complexatie is van cruciaal belang als unchelated Gd 3+ is giftig; Het is betrokken bij de ontwikkeling van nefrogene systemische fibrose bij sommige patiënten met nierziekte of mislukking 2. In het kader van nieuwe GBCA ontdekking in het onderzoek en de proof-of-concept niveau, arelatively gemakkelijker, sneller en kosteneffectieve spectroscopische gebaseerde methode (zoals UV-Vis absorptie of fluorescentie) kan dienen als een waardevol alternatief. Met deze toepassingen in het achterhoofd, een fluorescent-aptameergebaseerde sensor voor waterige Gd 3+ werd ontwikkeld 9. De aptameer (Gd-aptameer) is een 44-base lange enkelstrengs DNA-molecuul met een specifieke sequentie van basen die werd geïsoleerd door het proces van systematische evolutie van liganden door exponentiële verrijking (SELEX) 9. De aptameer te vormen tot een fluorescerende sensor, wordt een fluorofoor aan het 5&#39;-uiteinde van de streng, die vervolgens gehybridiseerd met een quenching streng (QS) via 13 complementaire basen (figuur 1) bevestigd. Het QS is gelabeld met een donkere quenchermolecuul op de 3 &#39;terminus. Aangezien Gd 3+, de sensor (Gd-sensor), bestaande uit een 1: 2 respectievelijk molverhouding van Gd-aptameer en QS, zal een minimale fluorescentie-emissie als gevolg to energieoverdracht van de fluorofoor naar de onderdrukker. De toevoeging van waterig Gd 3+ zal de QS verplaatsen van de Gd-aptameer, resulterend in een toename in fluorescentie-emissie. <img alt="Figuur 1" src="/files/ftp_upload/55216/55216fig1.jpg" /> Figuur 1. De sensor (Gd-sensor), die bestaat uit de 44-voet lange aptameer (Gd-aptameer) getagged met fluoresceïne (een fluorofoor) en de 13-voet lange blussen streng (QS) getagged met DABCYL (een donkere quencher) . Deze test maakt gebruik van het molecuul xylenol oranje, die een verschuiving van de maximale absorptiegolflengte 433-573 nm bij chelatie het ion 10 ondergaat. De verhouding van deze twee absorptiemaxima kan worden gebruikt om de hoeveelheid unchelated Gd 3+ kwantificeren. De aptameer sensor een alternatief (ook complementair zijn) aan de xylenol oranje assay, zoals de twee methoden hebben verschillende reactiecondities (zoals pH en samenstelling van de bufferoplossingen gebruikt), doel selectiviteit, lineaire reeksen van kwantificering en detectie voorwaarden 9.

A method for determining the presence of unchelated trivalent gadolinium ion (Gd3+) in aqueous solution is demonstrated. Gd3+ is often present in samples of gadolinium-based contrast agents as a result of incomplete reactions between the ligand and the ion, or as a dissociation product. Since the ion is toxic, its detection is of critical importance. Herein, the design and usage of an aptamer-based sensor (Gd-sensor) for Gd3+ are described. The sensor produces a fluorescence change in response to increasing concentrations of the ion, and has a limit of detection in the nanomolar range (~100 nM with a signal-to-noise ratio of 3). The assay may be run in an aqueous buffer at ambient pH (~7 - 7.4) in a 384-well microplate. The sensor is relatively unreactive toward other physiologically relevant metal ions such as sodium, potassium, and calcium ions, although it is not specific for Gd3+ over other trivalent lanthanides such as europium(III) and terbium(III). Nevertheless, the lanthanides are not commonly found in contrast agents or the biological systems, and the sensor may therefore be used to selectively determine unchelated Gd3+ in aqueous conditions.
