La nanomedicina está cambiando el paradigma del desarrollo del tratamiento del cáncer 1. Inspirado por el tremendo impacto clínico de cáncer nanomedicamentos anteriores, tales como liposomas y nanoterapias a base de albúmina 2, 3, muchas formulaciones novedosas se han producido en la última década. Sin embargo, los recientes análisis del éxito de la traducción clínica de estos nanomedicamentos cáncer indican que sólo unos pocos de ellos han sido aprobados para uso clínico 4, 5. Un obstáculo importante para la traducción clínica de nuevas nanomedicamentos cáncer es su limitada mejora del índice terapéutico en comparación con la administración directa de los compuestos terapéuticos gratuitos 6. Como tal evaluación, exacta del comportamiento in vivo de nanomedicamentos en sistémica, el tejido, y los niveles celulares en modelos animales preclínicos es esencial para identify aquellos con índices terapéuticos óptimos para futuro traducción clínica. Los nanomateriales pueden ser radiomarcados para la caracterización cuantitativa en animales vivos con tomografía por emisión de positrones (PET), que tiene excelente sensibilidad y reproducibilidad entre todas las modalidades de formación de imágenes clínicas 7. Por ejemplo, 89 Zr-etiquetado largo circulante nanomedicamentos se han caracterizado en modelos de ratón de cáncer de 8, 9, 10, así como en otros modelos de enfermedad 11. Además, la vida media en sangre y la biodistribución de los nanomedicamentos pueden ser evaluados ampliamente mediante el uso de mediciones ex vivo de radiactividad en los tejidos individuales 8. Por lo tanto, radiomarcaje permite la evaluación cuantitativa de nanomedicamentos en los niveles sistémicos y tejidos. Es importante destacar que, radiolabelenanomedicamentos d generalmente no se pueden analizar en el de una sola célula o los niveles subcelulares debido a la resolución espacial limitada de la señal radiactiva. Por lo tanto, el etiquetado fluorescente demuestra ser una modalidad complementaria para la evaluación de las nanopartículas con las técnicas de formación de imágenes ópticas tales como la citometría de flujo y microscopía de fluorescencia 12. Con este fin, las nanopartículas marcadas con radioisótopos y etiquetas fluorescentes se pueden evaluar cuantitativamente in vivo mediante formación de imágenes nuclear y ex vivo mediante recuento de radiactividad, y también pueden ser caracterizadas extensivamente a nivel celular por formación de imágenes ópticas. Anteriormente, hemos desarrollado procedimientos modulares para incorporar marcadores radiactivos y fluorescentes en varias nanopartículas, incluyendo lipoproteínas de alta densidad (HDL) 11, los liposomas 9, 10, las nanopartículas poliméricas, fragmentos de anticuerpos, y nanoemulsions 10, 13. Estas nanopartículas marcadas han permitido la caracterización cuantitativa en modelos animales pertinentes a distintos niveles, que han guiado a la optimización de estos nanomateriales para sus aplicaciones específicas. En el estudio actual, el objetivo es utilizar nanopartículas liposomales la plataforma de la nanomedicina más establecido 14 -como un ejemplo para demostrar procedimientos exhaustivos para generar una nanopartícula de doble etiquetado y caracterizar a fondo en un melanoma B16-F10 singénico clásico modelo de ratón 15 . A partir de los resultados, estamos seguros de este enfoque caracterización de nanopartículas se puede adaptar para evaluar otros nanomedicamentos cáncer en modelos de ratón pertinentes.

Inspirado por el éxito de nanomedicamentos cáncer anteriores en la clínica, los investigadores han generado un gran número de nuevas formulaciones en la última década. Sin embargo, sólo un pequeño número de nanomedicamentos han sido aprobados para uso clínico, mientras que la mayoría de nanomedicamentos en desarrollo clínico han producido resultados decepcionantes. Un obstáculo importante para el éxito de la traducción clínica de nuevas nanomedicamentos cáncer es la falta de una comprensión exacta de su actuación en vivo. En este artículo se dispone de un procedimiento riguroso para caracterizar el comportamiento in vivo de nanomedicamentos en ratones portadores de tumor en sistémica, de tejidos, de una sola célula, y los niveles subcelulares a través de la integración de emisión de positrones tomografía computarizada (PET-CT), los métodos de la radiactividad de cuantificación , citometría de flujo y microscopía de fluorescencia. Con este enfoque, los investigadores pueden evaluar con precisión nuevas formulaciones a nanoescala en modelos de ratón de signi pertinentesr. Estos protocolos pueden tener la capacidad de identificar los nanomedicamentos cáncer más prometedores con un alto potencial de traslación o para ayudar en la optimización de nanomedicamentos cáncer para la traducción futuro.

The poor understanding of the in vivo performance of nanomedicines stymies their clinical translation. Procedures to evaluate the in vivo behavior of cancer nanomedicines at systemic, tissue, single-cell, and subcellular levels in tumor-bearing immunocompetent mice are described here. This approach may help researchers to identify promising cancer nanomedicines for clinical translation.
