Her rapporterer vi fremgangsmåten for å utvikle en programmerbar substrat er i stand til å reagere på et kjemisk signal fra en manipulert cellelinje. 1 Vi gjør dette ved å opprette en biotin-streptavidin grensesnitt som svarer til biotin produsert av syntetisk konstruert Escherichia coli (E. coli) celler. Denne stamme ble utviklet ved hjelp av restriksjonsenzymkloningsfremgangsmåter for å sette sammen et plasmid, pKE1-lacl-bioB. Dette plasmid, når transformert inn i E. coli-stamme K-12 MG1655, begaver celler med evnen til å uttrykke høye nivåer av bioB, et essensielt enzym for biotin syntese. using synthetic biology to engineer living cells that interface with programmable materials

Vi har utviklet en abiotiske-biotiske grensesnitt som gjør det mulig modifiserte cellene for å kontrollere materialegenskapene til en funksjonalisert overflate. Dette systemet er fremstilt ved å lage to moduler: et syntetisk konstruert stamme av E. coli-celler og en funksjonalisert materiale grensesnitt. Innenfor denne artikkelen, vi detalj en protokoll for genetisk ingeniør utvalgte atferd innenfor en stamme av E. coli ved hjelp av molekylære kloning strategier. En gang utviklet, denne stammen produserer høye nivåer av biotin når de utsettes for en kjemisk inducer.
