Ice-bindingseiwitten (IBP) zijn een diverse familie van beschermende eiwitten die zijn ontdekt in een aantal organismen, waaronder planten 1, 2 insecten, vissen 3 en 4 microben. Het belangrijkste kenmerk van deze eiwitten is hun unieke mogelijkheid om gericht en efficiënt adsorberen aan ijskristallen, het wijzigen van hun groei. IBP meerdere gedocumenteerde eigenschappen, met de twee meest kenmerk dat thermische hysterese (TH) en ijs herkristallisatie inhibitie (IRI). TH activiteit gemakkelijker waargenomen IBP geproduceerd in freeze-intolerant dieren. Deze activiteit leidt tot verlaging van het vriespunt van de bloedsomloop of interstitiële vloeistoffen organismen om bevriezing te voorkomen. identification of plant ice-binding proteins through assessment of ice-recrystallization inhibition and isolation using ice-affinity purification

Ice-bindende eiwitten (IBP) behoren tot een familie van stress-geïnduceerde eiwitten die worden gesynthetiseerd door bepaalde organismen blootgesteld aan temperaturen onder nul. In planten vorstschade ontstaat wanneer extracellulaire ijskristallen groeien, wat resulteert in het breken van plasmamembranen en eventuele celdood. Adsorptie van IBP ijskristallen beperkt verdere groei door een proces dat ijs herkristallisatie inhibitie (IRI), waardoor celbeschadiging verminderen. IBP laten ook de mogelijkheid om het vriespunt van de oplossing beneden het evenwicht smeltpunt, een eigenschap bekend als thermische hysterese (Th) activiteit onderdrukken. Deze beschermende eigenschappen hebben belangstelling voor de identificatie van nieuwe IBP&#39;s verhoogd als gevolg van hun potentieel gebruik in industriële, medische en agrarische toepassingen. Dit document beschrijft de identificatie van planten IBP m 1) de inductie en extractie van IBP in plantenweefsel, 2) de screening van extracten van IRI activiteit en 3) het isoleren en purificatie van IBP&#39;s. Na de inductie van IBP door blootstelling lage temperatuur worden extracten getest op activiteit met behulp van een IRI &#39;splat assay&#39;, waarbij de observatie van de groei van ijskristallen met een standaard lichtmicroscoop maakt. Deze test vereist een lage eiwitconcentratie genereert resultaten verkregen die snel en gemakkelijk te interpreteren, verschaffen een eerste screening voor ijs bindingsactiviteit. IBP kan daarna worden geïsoleerd uit contaminerende eiwitten door gebruik van de eigenschap IBP adsorbeert ijs, via een techniek genaamd &quot;ice-affiniteitzuivering. Gebruik cellysaten vanuit plantenextracten, kan een ijs hemisfeer traag worden gekweekt op een koperen probe. Dit omvat IBP in de kristallijne structuur van de polykristallijne ijs. Waarbij geen a priori biochemische of structurele kennis van de IBP maakt deze werkwijze voor het terugwinnen van actief eiwit. -Ijs gezuiverde eiwitfracties kunnen worden gebruikt voor verdere toepassingen inclusief het bepalen van peptide sequenties door massaspectrometrie en biochemische analyse van natieve eiwitten.

Voor het gemak van de set-up, figuur 1 en Figuur 2 worden als visuele representaties van de voor IRI analyse en ijs-affiniteitzuivering respectievelijk materiaal. IRI resultaten van analyse met extracten vanuit mosterd onkruid met en zonder IBP zijn weergegeven in figuur 3.
